Clinical Significance Of MicroRNA-21and MicroRNA-205in Non-small Cell Lung Cancer

Objective: To investigate the expression level and clinical significance of microRNA-21(miRNA-21) and microRNA-205(miRNA-205) in non-small cell lung cancer (NSCLC)patients’ bronchoscopy tissue and plasma．Methods：To quantified the expression level of miRNA-21and miRNA-205in thirty oneNSCLC cases and ten controls with Real-time PCR，then compared the expression level ofeach microRNA between the two groups and analyzed correlation between the expressionlevel and clinicopathologic features of NSCLC patients for each microRNA．Meanwhile，we evaluated the diagnostic vaule of concerning microRNAs for NSCLC．Results:(1)The expression level of both miRNA-21and miRNA-205were significantlyupregulated in patients’ bronchoscopy tissue and plasma compared with controls (P＜0.05).(2) Overexpression of tissue miRNA-21was strongly associated with adenocarcinoma(AD)and advanced stage (stages III and Ⅳ) of NSCLC(P＜0.05)；The expression level ofplasma miRNA-21was higher in NSCLC patients with lymph node metastasis than thepatients without (P＜0.05) and the level in NSCLC patients with early stage(stages I and II)was also higher than the controls (P＜0.05)．(3) The tissue miRNA-205was obviouslyoverexpressed in squamous cell carcinoma (SCC) compared with AD(P＜0.05)．while thelevel of plasma miRNA-205had no association with clinicopathologic features．To besurprized，we found that the discrepancy of expreesion level of plasma miRNA-205between SCC and controls had statistic significance(P＜0.05)， while the statisticsignificance was not fund between AD and controls．(4)We applied ROC curve to evaluatethe miRNA-21diagnostic value for NSCLC in patients’ plasma and bronchoscopy tissue， both sensitivity and specificity were80.0%、70.0%；73.3%、80%respectively，andmiRNA-205got the sensitivity and specificity with81.3%、84.2%；81.3%、83.3%respectively．Conclusions：(1)Both miRNA-21and miRNA-205were overexpressed inbronchoscopy tissue and plasma of NSCLC patients．(2)MiRNA-21may have relationshipwith TNM staging in NSCLC patients，in addition，the tissue miRNA-21had thedifferential diagnostic value for AD and plasma miRNA-21could be considered as amarker for NSCLC screening．(3) MiRNA-205was hightly specific for SCC both inbronchoscopy tissue and plasma．(4) MiRNA-21and MiRNA-205could be considered asnovel tumor markers for NSCLC．...
