Objective: The results of the study were based on the correlation between levels of serum micro ribonucleic acid-128b(mi R-128b),micro ribonucleic acid-214(mi R-214)and biochemical indicators in Parkinson’s disease(PD)patients.This study is aimed to investigate the association between the levels of mi R-128 b,mi R-214 as well as biochemical indicators and severity and PD with depression(PD-D),and provide a theoretical basis for the study of PD biomarkers.Methods: A total of 60 patients with PD hospitalized in the Department of Neurology of Hebei Provincial General Hospital from December 1,2021 to October 31,2022 was selected as the PD group and50 healthy subjects from Physical Examination Center were selected as the control group.According to the Hoehn-Yahr staging(H-Y staging),PD patients was divided into mild group(H-Y staging from 1 to 2.5)and moderate to severe group(H-Y staging > 2.5).Furthermore,PD group was also divided into PD-D group(Hamiltion Depression Scale-24(HAMD-24)HAMD ≥ 8 points)and non-PD-D group(HAMD < 8 points)according to the HAMD-24.The expression levels of mi R-128 b and mi R-214 in serum were detected by real-time fluorescence quantitative polymerase chain reaction(q RT-PCR).General data and blood indicators were statistically analyzed,and the correlation between serum mi R-128 b,mi R-214 and uric acid(UA)levels and the severity of PD and PD-D was explored.Results: 1.In the PD group,33 cases were mild and 27 cases were moderate and severe.There were 39 cases in PD-D group and 21 cases in non-PD-D group.2.There was no significant difference in epidemiological data between PD group and control group(P > 0.05).3.The levels of serum mi R-128 b and serum UA in PD group were low,and the levels of serum mi R-214 was high(P < 0.05),compared with the control group.Multivariate Logistic regression analysis showed that serum mi R-128b(OR=0.327,95%CI: 0.085~1.252,P < 0.05)and UA(OR=0.998,95%CI:0.980~0.997,P < 0.05)were protective factors for PD,and serum mi R-214level(OR=1.629,95%CI: 1.119~2.373,P < 0.05)was an independent risk factor for PD.According to receiver operating characteristic curve(ROC)analysis,the serum mi R-128 b cut-off value was 2.100 with sensitivity of58.3% and specificity of 80.0%,with the area under the curve(AUC)of0.695(95%CI: 0.596~0.795,P < 0.05),the serum mi R-214 cut-off value was 30.192 with sensitivity of 80.0% and specificity of 66.0%,with the AUC)of 0.695(95%CI: 0.596~0.795,P < 0.05)as well as the serum UA cut-off value was 320.065mmol/L with sensitivity of 83.3% and specificity of 44.0%.4.According to H-Y staging,PD group was divided into mild group and moderate and severe group.The serum mi R-128 b level in mild group was higher than that in moderate and severe group,and the difference between the two groups was significant(P < 0.05).The serum mi R-214 level in mild group was lower than that in moderate and severe group.The difference between the two groups was significant(P < 0.05).5.According to HAMD-24 score,PD group was divided into PD-D and non-PD-D group.Compared with non-PD-D group,serum UA level in PD-D group was lower,and the difference between the two groups was significant(P < 0.05).6.Spearman correlation analysis showed that serum mi R-128 b level was negatively correlated with H-Y classification(r < 0,P< 0.05).Serum mi R-214 level was positively correlated with H-Y grading(r > 0,P < 0.05).Serum UA level was both negatively correlated with H-Y grade and PD-D(r < 0,P < 0.05).Conclusion: 1.The level of serum mi R-128 b,mi R-214 and UA have a certain clinical value in the diagnosis of PD,in order to provide more biomarkers for the early clinical diagnosis of PD.2.Serum mi R-128 b,mi R-214 and UA levels are correlated with the severity of PD,which may provide help for clinical assessment of PD staging.3.PD patients with low levels of serum UA are more likely to be accompanied by depression.It is expected to provide new ideas for clinical exploration of risk factors for PD-D. |
