| Objective: The expression of miR-126-3p,miR-199a-3p,miR-150-5p and miR-221-3p in serum of patients with cervical cancer,patients with cervical precancerous lesions and normal physical examination subjects were detected.To explore the value and clinical significance of miRNA in the early screening of cervical cancer,and to provide some ideas for treatment and prognosis evaluation.Method: Patients hospitalized in our hospital from December 2020 to December 2022 were selected,61 female patients diagnosed with cervical cancer by pathology were selected as CC group,40 female patients diagnosed with high-grade squamous intraepithelial lesions by pathology were selected as HSIL group,and 40 healthy women who underwent physical examination during the same period were selected as control group.The relative expression of miR-126-3p,miR-199a-3p,miR-150-5p and miR-221-3p in blood samples of the three groups of subjects was detected by RT-q PCR.In the CC group,42 patients underwent surgical treatment(Modified extensive total hysterectomy plus pelvic lymph node ±abdominal aortic lymph node dissection),and the relative expression levels of each miRNA in serum were re-examined 7 days after surgery.Results: 1.The relative expression level of miR-126-3p in serum of patients in control group,group HSIL and group CC was gradually decreased,and the difference was statistically significant(P<0.05);The relative expression level of miR-199a-3p in serum of CC group was lower than that of control group and HSIL group,and the difference was statistically significant(P<0.05).The relative expression level of miR-150-5p in serum of patients in control group,group HSIL and group CC was gradually increased,and the difference was statistically significant(P<0.05).The relative expression level of miR-221-3p in serum of CC group was higher than that of control group and HSIL group,and the difference was statistically significant(P<0.05).2.The relationship between each miRNA and the clinical characteristics of patients in the CC group was analyzed,and the results showed that the relative expression level of miR-126-3p in the serum of patients in the CC group was correlated with the FIGO stage and the degree of tumor differentiation(P<0.05).The later the FIGO stage,the worse the tumor differentiation and the lower the relative expression level.The relative expression level of miR-199a-3p was correlated with vascular invasion(P<0.05),and the relative expression level of miR-199a-3p in patients with vascular invasion was lower than that in patients without vascular invasion.The relative expression level of miR-150-5p was correlated with lymph node metastasis(P<0.05),and the relative expression level of miR-150-5p was higher in patients with lymph node metastasis than those without lymph node metastasis.The relative expression level of miR-221-3p was correlated with FIGO stage and lymph node metastasis(P<0.05).The later the FIGO stage was,the higher the relative expression level of miR-221-3p was in patients with lymph node metastasis.3.According to ROC curve analysis,the sensitivity of serum miR-126-3p,miR-199a-3p,miR-150-5p and miR-221-3p in the diagnosis of cervical cancer were 93.3%,91.7%,85.0% and 81.7%,respectively.The specificity was 70.0%,82.5%,83.7%,80.0%,and the area under the curve was 0.865,0.874,0.913,0.876,respectively.The optimal critical point for diagnosis was 0.40,2.91,2.50,2.49,respectively.The detection sensitivity,specificity and AUC of miR-126-3p combined with miR-199a-3p were95%,76.3% and 0.903,and the detection sensitivity,specificity and AUC of miR-150-5p combined with miR-221-3p were 91.7%,85.0% and 0.941.4.The relative expression levels of miR-126-3p and miR-199a-3p in serum of patients in CC group were increased after surgery,and the difference was statistically significant(P<0.05);The relative expression levels of miR-150-5p and miR-221-3p in serum after surgery were decreased,and the difference was statistically significant(P<0.05).Conclusion:1.miR-126-3p,miR-199a-3p,miR-150-5p and miR-221-3p are abnormally expressed in the sera of patients with cervical c ancer and precancerous lesions,which are closely related to the occur rence and development of cervical cancer and may provide a new tar get for the treatment of cervical cancer.2.The relative expression level of miR-126-3p in serum of CC group was correlated with FIGO stage and differentiation level;The relative expression level of miR-199a-3p in CC group was correlated with vascular infiltration.The relative expression level of miR-150-5p in serum of CC group was correlated with lymph node metastasis.The relative expression level of miR-221-3p in serum of CC group was correlated with FIGO stage and lymph node metastasis.It may be related to the progression and prognosis of cervical cancer.3.The detection of miR-150-5p combined with miR-221-3p showed high sensitivity and specificity in the screening of cervical cancer,and could be considered as a marker for the early screening of cervical cancer.4.The differential expression of miR-126-3p,miR-199a-3p,miR-150-5p and miR-221-3p in serum of patients in the CC group after s urgery provided a certain reference for the prognosis assessment of p atients with cervical cancer. |
