Study On The Quality Characteristics And Specific Barcodes Of Rhubarb From Different Authentic Production Areas Based On LC-MS And Chloroplast Gene Sequence

Rhubarb is a common herb in bulk with the increase of the dosage of medicine,rhubarb,wild resource overexploitation serious,and some real estate areas in five in the process of introduction of wild confusion,geoherbalism germplasm resources have been lost,urgent to carry out the evaluation of the genuineness of germplasm resources work.The material basis of the pharmacological action of rhubarb is a variety of functional components,and the components of its efficacy change with the geographical and genetic factors.In order to improve the clinical curative effect of rhubarb,it is necessary to make a deep analysis on the quality of Rhubarb in different areas,in order to divide the special medicinal materials according to the producing area and the effective components.According to the environmental conditions and the origin of ancient literature records on the real estate area within the division of the estate area;in different areas rhubarb samples collected earlier research group as research material,matK sequence analysis of different haplotypes and estate rhubarb based on distribution characteristics,study the identification efficiency of different real estate rhubarb,for identification can not be carried out through the matK sample,can choose the sequence selected from chloroplast genome level;in addition,the LC-MS of 70 questionnaires collected from different areas of big yellow estate root and stem samples of 24 kinds of chemical constituents were determined at the same time,in accordance with the "diarrhea attack plot,heat purging through the fire,stasis" effect of component content difference of the quality characteristics of different real estate areas of medicine analysis.The results are as follows:1 Combined with the records of the real estate and environmental conditions,such as the production of large areas of Sichuan real estate division:Qinghai area produced "Xining rhubarb",produced in the region,"Ya huang".Gansu and Lanzhou counties to the northwest and produced "Liangzhou rhubarb" in Dingxi and neighboring Lintan,Zhuoni,Minxian region,Minxian,Linxia,Gannan,rhubarb,produced by "Hezhou rhubarb" in South Minxian,Lixian,Zhouqu,Dangchang County,Wenxian and the west,near the late the "Ambrose rhubarb" Tianshui,produced by "water rhubarb" in the vicinity of the Pingliang produced"Zhuanglang rhubarb".2 The whole genome sequence of different producing area of matK gene and rhubarb rhubarb samples were analyzed in the chloroplast:Rhubarb matK haplotype the same real estate areas are similar,at the same time in the system clustering also clustered into one,this genetic mechanism may cause the differences of Rhubarb in different real estate areas of quality,but also found some haplotypes there are a number of real estate in the area,indicating different areas rhubarb gene flow may exist,therefore,only through the matK haplotype to different real estate areas rhubarb identification.Analysis of tandem repeat sequences and SNP/InDel of the whole genome sequence of chloroplast rhubarb samples,in order to obtain the molecular identification of Rhubarb in different estate interval;comparative analysis of the complete genome sequence of Rheum palmatum and medicinal rhubarb chloroplast,screening high variation zone ndhE-ndhG,ndhG-ndhI,rp12-psbA sequence as the potential for the identification of bar code,identification efficiency,need further studies with larger sample volume.3 The different real estate area effect component content of rhubarb Rhubarb in different studies:real estate areas in the "diarrhea attack plot,clearing heat and purging fire,removing blood stasis and effect group significantly different content,Hezhou components by rhubarb by silt content is the highest,can be used as medicinal materials through the blood stasis type potential;Huang Ya purging and product composition was the highest,and can be used as potential purging and product special medicinal materials;Qingrexiehuo cool yellow efficacy component content is the highest,can be used as a potential Qingrexiehuo special medicine.Rhubarb in different real estate areas rich in differentiation,different effects of component content and proportion in Chinese medicine in the future therefore,according to the function demand,respectively,different selections of real estate areas of medicine,in order to achieve a more accurate symptomatic medication,improve the curative effect of traditional Chinese medicine.The above analysis,the matK sequence to complete the identification of different real estate area based on the needs of rhubarb,chloroplast genome screening level resolution is more fragments;there are significant differences in different quality of rhubarb real estate areas,these differences lead to its potential advantages in the functional components.This study provides basic technical research in different areas rhubarb estate support bar code identification,and suitable for clinical efficacy of rhubarb,rhubarb to ensure precise clinical drug and improve the curative effect of traditional Chinese medicine to provide the experimental basis for.At the same time,it also provides important theoretical basis for the quality oriented cultivation of rhubarb.
