| Objectives:Making use of nuclear magnetic resonance(NMR)metabolomics technology,to explore the characteristics of metabolite spectrum and the characteristics of metabolic markers of Allergic constitution;To further improve the Allergic constitution classification basis and formation mechanism of the experimental research,to provide theoretical and experimental basis for the clinical diagnosis and treatment,to better help for a precise judgement of Allergic constitution,also to explore and enrich the TCM Constitution classification Content and its formation mechanism accordingly.In recent years,immunological studies have shown that Allergic constitution and allergic diseases are closely related to the immune function of the body.On the basis of obtaining the potential biomarkers of Allergic constitution,this study combines immunology to analyze the relationship between potential biomarkers of Allergic constitution and its metabolic pathways and immune function,and tries to find the relationship between them.Methods:This study was based on the theory of constitution of TCM and was supported by previous theoretical research,experimental research,and clinical research.All objects of study were provided by the China-Japan Friendship Hospital Medical Examination Center and the Beijing Hongyitang Traditional Chinese Medicine Hospital.To identify Constitution,researchers through the combination of TCM Constitution Scale identification and manual identification,Allergic constitution and normal constitution according to the"TCM Constitution classification and determination criteria"(Chinese Society of Traditional Chinese Medicine:ZZYXH/T157-2009)to determine(attachment Table),eventually filtered into groups and divide them into two groups,namely Allergic constitution group and normal constitution group(control group),30 cases in each group,a total of 60 cases.Then researchers collected blood and urine samples at the same time and place,since sample bening processed,researchers utilized the Varian(Varian,Inc.)INOVA 600 MHz superconducting NMR spectrometer and the 1H-NMR technology,detected and analyzed the blood and urine metabolite spectrum characteristics of two groups to further seek for the potential biomarkers of Allergic constitution.Result:Allergic constitution have a metabolic network change in comparison with normal constitution,and it has been discovered potential biomarkers on Allergic constitution and has been defined the direction of their content changes.After a review of the literature,we found that among the potential biomarkers that are different,there are 6 potential biomarkers related to the activity of immune regulation.They are lipoprotein,glucose,N-acetyl glycoprotein,acetic acid,alanine and glycine.They involve lipid metabolism,glucose metabolism,energy metabolism and amino acid metabolism.In the urine samples of the Allergic constitution group,6 potential biomarkers were found,namely citrate(δ2.54,2.66-2.70),3-hydroxybutyrate(δ1.18,1.22),creatinine(δ3.06,4.06),creatine(δ3.94),glucose(δ3-4),TMAO(δ3.26).The potential biomarkers in urine concerning Allergic constitution was that 3-hydroxybutyrate(δ1.18,1.22),TMAO(δ3.26),creatinine(δ3.06,4.06)were lower than the normal constitution and glucose(δ3-4),citrate(δ2.54,2.66-2.70),creatine(δ3.94)were higher than the normal constitution.In the blood samples of the Allergic constitution group,8 potential biomarkers were found,namely Lipoprotein(δ0.82-0.88,1.25-1.30),Glycine(δ3.55-3.56),alanine(δ1.46-1.49),Acetic acid(δ1.91-1.92),NAc(δ2.02-2.05),glucose(δ3.20-4.00),3-hydroxybutyrate(δ1.19-1.20),Citrate(δ2.50-2.56,2.64-2.70).The potential biomarkers in blood concerning Allergic constitution was that Citrate(δ2.50-2.56,2.64-2.70),3-hydroxybutyrate(δ1.19-1.20),Acetic acid(δ1.91-1.92),NAc(δ2.02-2.05),Lipoprotein(δ0.82-0.88,1.25-1.30)were lower than the normal constitution and alanine(δ1.46-1.49),glucose(δ.20-4.00),Glycine(δ3.55-3.56)were higher than the normal constitution.Conclusions:This study has discovered the potential biomarkers of the Allergic constitution,and made clear the direction of their content changes.It is to be found that the Allergic constitution is not only abnormal in lipid metabolism,sugar metabolism,amino acid metabolism and energy metabolism,but also the changes of neurotransmitters and viscera function.We found that among the potential biomarkers that are different,there are 6 potential biomarkers related to the activity of immune regulation.They are lipoprotein,glucose,N-acetyl glycoprotein,acetic acid,alanine and glycine.They involve lipid metabolism,glucose metabolism,energy metabolism and amino acid metabolism.This study illustrates that the application of Metabolomics method can objectively identify Allergic constitution and provide experimental evidence for TCM Constitution classification. |
