Objective:With the increasing incidence of ulcerative colitis(ulcerative colitis,UC)in recent years,it has become one of the refractory diseases recognized by the World Health Organization(WHO).At present,the treatment of UC is mainly induced remission in active stage and maintenance remission in remission stage,but UC is easy to recur in remission stage,and it is often repeated,so it is difficult to achieve clinical complete recovery is an important problem in the treatment of UC.At present,a large number of studies have confirmed that the degree of intestinal flora imbalance is related to the stage and activity of the disease,and the main pathogenesis of UC in the remission stage of spleen deficiency has been confirmed by a large number of studies.Therefore,the main purpose of this study was to determine the changes of intestinal flora in patients with UC spleen deficiency syndrome in remission stage and healthy people.In order to explore the mechanism of recurrence and recurrence of UC in remission stage from the point of view of intestinal flora and to explore the correlation between UC and intestinal flora of spleen deficiency syndrome.Methods:A total of 32 patients with ulcerative colitis with spleen deficiency syndrome in remission stage were collected from the Department of Gastroenterology,Oriental Hospital of Beijing University of traditional Chinese Medicine.Twenty-nine healthy volunteers from Beijing,Chongqing,Henan,Ningxia and other places were collected.The fecal samples of patients and volunteers were collected,and the total NDA,was extracted by 16SrDNA technique.the total DNA was detected by Illumina MiSeq,the database was established,and the species analysis was carried out.the difference of intestinal flora between UC spleen deficiency syndrome in remission stage and healthy people was obtained.Results:There is intestinal flora imbalance in spleen deficiency syndrome of ulcerative colitis in remission stage.The diversity of intestinal flora in ulcerative colitis is significantly lower than that in healthy group.there is a significant difference in the structure of intestinal flora between the two groups.Compared with the healthy group,the dominant flora in feces of UC patients with spleen deficiency syndrome in remission stage were as follows:1.phylum level:Proteobacteria、Saccharibacteria;2.class level:No significant difference in flora;3.order class:Aeromonadales、Neisseriales、Pseudomonadales、Actinomycetales;4.family level:Bacteroidaceae、Lachnospiraceae、Aeromonadaceae、Neisseriaceae、Camobacteriaceae、Moraxellaceae、Actinomycetaceae。5.genus level:Bacteroides、Ruminococcus-gnavusgroup、Lachnoclostridium、Blautia、Anaerostipes、Megasphaera、RuminococcaceaeUCG-009、Oribacterium、Aeromonas、Citrobacter、Peptostreptococcus、Flavonifractor、Holdemanella、Neisseria、ErysielotrichaceaeUCG-007、Acinetobacter、Enterobacter、Coprococcus1、Erysipelatoclostridium、Prevotella、Prevotella7、Actinomyces。Compared with the healthy group,the abundance of UC spleen deficiency group in remission stage was lower as following:1.phylum level:Firmicutes、Fusobacteria;2.class level:Fusobacteriia、Deltaproteobacteria;3.order class:Fu-sobacteriales、Desulfovibrionales;4.family level:Prevotellaceae、Ruminococcac-eae、Rikenellaceae、Acidaminococcaceae、Christensenellaceae、FamilyXI、Desu-lfovibrionaceae、ClostridialesvadinBB60group;5.genus level:Subdoligranulum、Alistipes、LachnospiraceaeUCG-010、Ruminococcus1、RuminococcaceaeUCG-002、Eubacterium-ruminantium group、Ruminococcus2、Phascolarctobacterium、Ch-ristensenellaceaeR-7group、Eubacterium-ruminantiumgroup、RuminococcaceaeUC-G-014、LachnospiraceaeNK4A136group、Coprococcus2、RuminococcaceaeUCG-003、LachnospiraceaeND3007group、LachnospiraceaeFCS020group、FamilyXIIIAD3011 is intestinal flora imbalance in spleen deficiency syndrome of ulcerative colitis in remission stage.The diversity of intestinal flora in ulcerative colitis is significantly lower than that in healthy group.there is a significant difference in the structure of intestinal flora between the two groups.②The spleen deficiency syndrome of UC in remission stage was more obvious than that in healthy group in Bacteroides,Spirulaceae,Proteus,Ruminococcus-gnavusgroup and Lachnoclostridium.These fungi may produce virulence factors related to UC,which may lead to intestinal epithelial dysfunction,affect intestinal mucosal barrier function,so that intestinal mucosa can not fully heal,resulting in recurrent attacks in patients with UC spleen deficiency syndrome in remission stage.③ At the same time,the decrease of UC spleen deficiency syndrome was most obvious in Prevo’s family,rumen coccidiosis,thick-walled bacteria,Subdoligranulum,scientific research bacteria,LachnospiraceaeUCG-010,Ruminococcusl and RuminococcaceaeUCG-002.It is suggested that the decrease of the above flora abundance may lead to the decrease of intestinal immune function in UC spleen deficiency group in remission stage,so that can not play its protective effect on intestinal tract,but the important reason for the recurrence and delay of UC spleen deficiency syndrome in remission stage.
