Effect Of Qufeng Ningkui Recipe On The Intestinal Flora Of Patients With Ulcerative Colitis And Large Intestine Damp-heat Syndrome

Objective:To investigate the effect of Qufeng Ningkui Decoction on the intestinal flora of the large intestine damp-heat syndrome in the active period of ulcerative colitis.Methods:This study will meet the inclusion criteria of 20 patients with ulcerative colitis active large intestine damp-heat syndrome,by oral administration of Qufeng Ningkui Decoction.The clinical efficacy and changes of intestinal flora before and after treatment were evaluated after 4 weeks of treatment.Another healthy control group was used for the detection of intestinal flora in 8 healthy volunteers who were recruited publicly,and the difference in intestinal flora between UC patients and healthy people was compared.Results:A total of 19 patients with ulcerative colitis active large intestine dampness syndrome and 8 healthy volunteers completed a case study.?According to the evaluation of the overall score of TCM syndrome,6 cases were markedly effective in 19 cases,accounting for 32%of the total.10 cases were effective,the total effective rate was 84.2%,and 3 cases were ineffective,accounting for 16%of the total.?After treatment,the endoscopic score,lesion range,main symptom score and mayo activity index of patients with UC were lo,wer than before.Except for the lesion range,the other evaluation indexes were statistically significant by rank sum test P<0.05,suggesting that after treatment,UC The condition is less severe than before.?A total of 2,340,112 optimized sequences were obtained by high-throughput sequencing technology,and clustered with 97%similarity,a total of 2321 OTUs were obtained.Alpha diversity analysis:Compared with the healthy control group,the Shannon index decreased and Simpson increased,suggesting that the sample diversity of the pre-treatment group was significantly lower than that of healthy people(P<0.05).The post-treatment group was more obvious than the pre-treatment Simpson.The decrease(P<0.05)and the Shannon index increased(P>0.05),suggesting that the sample diversity of the group increased after treatment.PLS-DA analysis:There were significant differences in the composition of UC patients and healthy people before and after treatment.The composition of different groups of UC patients was different.?At the level of gates,compared with the healthy control group,the abundance of Mycelia and Bacteroides in the pre-treatment group was significantly decreased(P<0.05),and the abundance of the acid bacteria was increased(P<0.05).Compared with the pre-treatment group,the abundance of actinomycetes and Bacteroides gates increased significantly after treatment(P<0.05),and the abundance of proteobacteria decreased significantly(P<0.05).?genus level:Pseudomonas,Bacteroides,Escherichia,Ruminococcus,Bacterial,Dorea,and Genus were significantly lower than the healthy control group before treatment(P<0.05),the genus Ralstonia,Enterococcus was significantly higher than the healthy control group(P<0.05).After treatment,the group of Bacteroides,Staphylococcus,Fusobacterium,Tyzzerella and S.cerevisiae increased significantly(P<0.05).?LEfSe multi-level species discriminant analysis:A total of 61 taxa were found in the healthy control group.A total of 5 taxa were found before treatment.A total of 38 taxa were found after treatment.Conclusion:The treatment of patients with ulcerative colitis in the active period of large intestine damp-heat syndrome is effective,and can significantly improve abdominal pain,diarrhea,mucus pus and bloody stools.The diversity of intestinal flora in UC patients is significantly lower than that in healthy people.Qufeng Ningkui Decoction may increase the abundance of intestinal probiotics,reduce the abundance of harmful bacteria,repair intestinal mucosal barrier,and regulate intestinal microecological balance.Achieve the role of treatment of UC.At present,there are limited types of microbial preparations that can be used in clinical practice.More and more studies have shown that Chinese herbal medicine can increase the diversity of intestinal bacteria and promote the ecological balance of the intestinal tract.Therefore,Qufeng Ningkui Decoction can be combined with microbial preparations to treat UC large intestine dampness syndrome.Patients,providing new ideas for clinical medication,deserve further study.
