Objective: To explore the effect of chemokine CXCL12/CXCR4 axis on angiogenesis after cerebral infarction and the intervention effect of Huoxue Rongluo recipe in vitro.Methods: The method of oxygen-glucose stripping / reoxygenation and reoxygenation was used to injure the BMECs of P3 rats in order to simulate the pathophysiological process of cerebral infarction.The drug-containing serum of Huoxue Rongluo prescription was prepared to interfere with the injured rat BMECs.Butylphthalide was used as the positive control drug,and AMD3100 inhibited CXCR4.The experiment was divided into five groups: normal control group,model group,butylphthalide group(OGD/R damage + butylphthalide intervention),Huoxue Rongluo prescription group(OGD/R damage + Huoxue Rongluo recipe intervention),inhibitor group(OGD/R damage + Huoxue Rongluo recipe intervention + AMD3100).CCK-8,scratch,transwell migration,and tube information test were used to detect the angiogenesis phenotype of BMECs after injuries,such as proliferation rate,scratch healing rate,migration number,and tube length.The protein expression of CXCL12,CXCR4,and VEGFA was detected by Western blotting,and the m RNA level of CXCL12 and CXCR4 was detected by the RT-q PCR method.Results: 1.The proliferation rate,scratch healing rate,cell migration number,and tube length phenotype of BMECs in Huoxue Rongluo recipe group were significantly higher than those in the model group.The proliferation rate,scratch healing rate,cell migration number,and tube length phenotype of BMECs in the inhibitor group were significantly lower than those in the Huoxue Rongluo prescription group.2.The protein expression of CXCL12,CXCR4,and VEGFA in Huoxue Rongluo prescription group increased significantly.In contrast,the expression of CXCR4 and VEGFA protein in BMECs in the inhibitor group was significantly lower than that in Huoxue Rongluo prescription group.CXCL12 and CXCR4 m RNA in the Huoxue Rongluo recipe group were significantly increased,and the expression of CXCR4 and m RNA in BMECs in the inhibitor group was significantly lower than that in Huoxue Rongluo prescription group.Conclusion: Huoxue Rongluo recipe can promote the angiogenesis of rat BMECs cells after OGD/R by promoting the expression of CXCL12/CXCR4 axis and activating VEGFA.
