| Objective: to establish the osteoporosis model of ovariectomized rats,detect the femoral bone mineral density and serum OPG,RANK and RANKL levels of ovariectomized rats,and verify the intervention effect of Bushen Huoxue Recipe on postmenopausal osteoporosis and whether it can prevent and treat postmenopausal osteoporosis through opg/rank/rankl signal pathway.It provides a more solid theoretical basis and scientific basis for the development of traditional Chinese medicine resources and its wide application in clinical practice.Methods: 32 3-month-old female SD rats were randomly divided into four groups:Traditional Chinese medicine group,estrogen group,model group and blank control group,with 8 rats in each group;The ovariectomized rat model was established.Two months after the model was established,the traditional Chinese medicine group was given Bushen Huoxue Decoction by gavage,the estrogen group was given the same volume of conjugated estrogen tablets solution,the model group and the blank group were given the same volume of 0.9% normal saline by gavage once a day for three months.Three months after intragastric administration,the samples were taken to detect the levels of serum OPG,rank and RANKL by ELISA,and the bone mineral density of the proximal femur of the right side of the rats by DXA.The results were statistically analyzed.Results:1 Bone mineral density: the bone mineral density of rats in blank group was higher than that of rats in traditional Chinese medicine group,estrogen group and model group(P;The BMD of the model group was lower than that of the traditional Chinese medicine group and the estrogen group(P < 0.01),while the BMD of the traditional Chinese medicine group was 0.722 compared with that of the estrogen group(P > 0.05),suggesting that the two groups have the same curative effect on postmenopausal osteoporosis.2.OPG: the serum OPG levels of rats in the blank group,the traditional Chinese medicine group and the estrogen group were higher than those in the model group,and the difference was statistically significant(P < 0.01).The serum OPG levels of rats in the traditional Chinese medicine group and the estrogen group were higher than those in the blank group,and the difference was statistically significant(P <0.01).However,there was no significant difference in OPG levels between the traditional Chinese medicine group and the estrogen group(P > 0.05).3.rank: the rank level of rats in the blank group,the traditional Chinese medicine group and the estrogen group was lower than that in the model group,and the difference was statistically significant(P < 0.01),while there was no significant difference in the rank level of the traditional Chinese medicine group,the estrogen group and the blank group(P > 0.05).4.rankl: the level of RANKL in the blank group,the traditional Chinese medicine group and the estrogen group was lower than that in the model group,and the difference was statistically significant(P < 0.01).The level of RANKL in the traditional Chinese medicine group and the estrogen group was lower than that in the blank group,and the difference was statistically significant(P < 0.01).However,there was no significant difference between the traditional Chinese medicine group and the estrogen group(P > 0.05).Conclusion: 1.Bushen Huoxue recipe has certain curative effect on prevention and treatment of postmenopausal osteoporosis.2.Bushen Huoxue recipe can promote the expression of OPG,inhibit the expression of rank and RANKL,and promote bone formation through the regulation of opg/rank/rankl system,so as to prevent and treat postmenopausal osteoporosis.3.the change of bone mineral density was positively correlated with serum OPG level and negatively correlated with serum rank and RANKL levels. |
