| Objective: This experiment was designed to observe the general situation of collagen-induced arthritis(CIA)model rats,serum interleukin-6(1L-6),β-catenin concentration differences,and WNT signaling pathway related proteins β-catenin,Wnt3 a,1L-6,MMP13 protein expression changes in the ankle tissue of rats,to explore the extent of the effect of Coix lacrymae ester on bone destruction and joint inflammatory changes in CIA rats,and to provide a new theoretical reference for the clinical treatment of rheumatoid arthritis.Materials and Methods.1.Molding and groupingFifty 6-week-old SD rats(SPF grade)were fed adaptive for one week,and 8 rats were randomly selected as blank group.The remaining 42 rats were modeled by CIA.The rats with successful modeling were randomly divided into 5 groups: CIA model group,Coix lactate low-dose group(2.1g/kg/d,KLT1),Coix lactate medium-dose group(4.2g/kg/d,KLT2),Coix lactate high-dose group(8.4g/kg/d,KLT4),methotrexate group(0.5mg/kg/w,MTX).There are 6 patients in each group.The remaining rats failed to be modeled and were not added to the experimental control.2.Index testExcept for the blank group,all the groups were administered from the first day after secondary immunization,and the general indexes of rats were measured(foot and paw swelling,arthritis index,systemic inflammation index),after 5 weeks of continuous administration,the patients were put to death under anesthesia.The serum levels of 1L-6 and β-catenin in each group were detected by ELISA,and the expression of WNT signaling pathway related proteins β-catenin and WNT3 a and the concentration changes of 1L-6 and MMP13 in ankle joints of rats were detected by WB.HE,crosin O solid green staining and TRAP staining were performed on the ankle joint pathology,and the relevant pathological indicators such as synovial hyperplasia,inflammatory cell infiltration,cartilage destruction and the number of osteoclasts were scored,and the pathological changes of Coix seed on CIA model mice were analyzed.Results.1: Compared with the blank group,the paw swelling and AL score of rats in model group were significantly increased(p<0.05),the paw swelling degree and AI score of CIA rats were significantly improved after the administration of Coix lacrymae ester(p<0.05),among which the high dose group(KLT4)of Coix lacrymae ester had the best effect.2: Compared with the CIA group,after the administration of different doses of Coix lacrymae ester,the synovial membrane proliferation,and inflammatory cell infiltration were significantly controlled,and cartilage and bone damage were improved in CIA model mice(p<0.05),and Coix lacrymae ester could improve ankle joint pathology.3: Compared with the blank group,the expression levels of 1L-6 andβ-catenin in the serum of the model group were significantly increased,and the expression of 1L-6 and β-catenin in the serum was decreased after administration of the drug compared with the model group,with the most significant trend of decrease in the high dose group of Coixin ester(p<0.05);compared with the blank group,the expression of β-catenin,wnt3 a,WNT3A in the ankle joint of the model group rats was Compared with the blank group,the protein expression of 1L-6,WNT3 A and MMP13 in the ankle joint of the model group was increased(p<0.05),and compared with the model group,the expression of 1L-6,β-catenin,WNT3 A and MMP13 showed a decreasing trend after different doses of C.cochleariae ester administration,and the decreasing trend was most obvious in the high dose group of C.cochleariae ester(p<0.05).Conclusion.1.Coix lacrymae ester inhibited the inflammatory response of joints in CIA rats2.Coix lacrymae ester can reduce bone damage in CIA rats3.Cochlearyl ester can inhibit the activation of wnt/β-catenin signaling pathway by suppressing the expression of 1L-6 and MMP13,thus achieving the repair of bone damage... |
