Immune milk is the milk that contains specific antibodies,which is the result of hyperimmunization of cows vaccined with selected antigens.Beside of milk antibodies,immune milk also contains various lymphokines induced by immune response,which can be transferred into milk during lactation.Anti-inflammatory factor in immune milk is one of these lymphokines. This research subject is to prepare immune milk and to study the properties of anti-inflammatory factor(AIF) in immune milk.Our research results are as follow: 24 strains of pathogenic bacteria (including 12 of pathogenic E. coli, 8 of Salmonelia,3 of Shigella and 1 of Yersinia enterocoiitica) originated from human intestinal tract were selected as antigens,and dairy cows were immunized.Vaccination could not increase the concentration of IgG in the immune milk,but significantly highten the specificity of mik antibodies.The antibody titers against 24 pathogenic bacteria in immune colostrum (20mg/mI of IgG) were 2~2 I2,32~256 folds of control regular colostrum (20mg/mi of IgG). The antibody titers against 24 pathogenic bacteria in immune milk were 2528,8~l28 folds of control regular milk. Comparing to the IgG isolated from regular colostrum,the specific IgG isolated from immune colostrum could significantly inhibit the growth of E.coli and Salmonella in vitro ,and could protect mice from diarrhea challenged with E. coil and Salmonella. The numbers of b~fidobactriwn and lactobacilius,and contents of lactic acid and acetic acid in intestinal tract of mice fed with immune milk powder were significantly higher than that of mice fed with regular milk powder.At the same time, the numbers of Enterrococcus and E. coil in intestinal tract of mice fed with immune milk powder were significantly lower than that of mice fed with regular milk powder.This is probably attributed to intestinal local immune regulation of specific antibodies and functions of lymphokines in immune milk. Feeding of immune milk powder could improve the immune functions of mice,and also could inhibit odema of rat footpad induced by carrageenin and formaldehyde and the formation of acestoma of mice caused by cotton embedding hypophloeodally.Meanwhile,the PGE2 level in inflammatory footpad of rat fed with immune milk powder was remarkably lower than that of rat fed with regular milk powder.These results indicated that immune milk powder has anti-inflammatory property. LD50 for mice feeding of immune infant milk formula (100mg/I OOg of IgG) prepared by adding of immune colostrum powder into infant milk formula was higher than I Og/kg. No aberration-inducing effects of immune infant milk formula were observed resulting from the 3 20O1~5~ Ames test~ mouse marrow micronucleus test and mouse sperm aberration test. 30-day feeding of immune infant milk formula showed that there is no side effects to mice,blood biochemical examinations were normal. Pathobiological inspection indicated that no pathobiological changes is occurred to the mice. Within the range of PH4.0-1O.0,JgG was very stable,PH<4.0 or PH>l0.0,the stability of lgG was decreased. IgG could resist to I .OM GuHCI. When the concentration of GuHC1 was higher than I .OM, the stability of JgG was declined. Sucrose exerted protection effect on the lgG denaturation induced by acid alkali heat and GuHCI.
