| Objective: To immunize cows with Helicobacter pylori meanwhile providing with qingrehuashi decoction and to acquire healthy immune milk which riches in Helicobacter pylori polyclonalantibody and traditional Chinese medicine active component.The ultimate result of this experiment is to prevent and treat the gastropathy of Helicobacter pylori.Method:(1) To immunize cows with Helicobacter pylori: three healthy cows were immunized by injecting Hp liquid per 2 ml .Before every immune and on the eighth day,the thirtieth day after the last immune collected the jugular vein blood and milk (to make serum).Antibody of the milk was determined individually by direct agglutination test and double agar diffusion .(2) To feed cows with traditional Chinese Compound and to detecte the ingredient of the traditional Chinese medicine in milk: the cows were fed with traditional Chinese Compound on seventh day of the last immune in the morning and at afternoon and at night. The jugular vein blood and all milk were collected after 12 hours of giving the first drug. At the same time detected the composition contained in Chinese medicine herbs decoction,Serum and milk by the methods of specific antibody test and double laminar analysis diffusion.(3) Experimental research on inhibitive action on Helicobacter pylori with the healthy immune milk in vitro: 5mm card was separately immersed in healthy immune milk,antibody milk,Chinese medicine milk,Chinese medicine decoction,milk and normal saline for 60 minutes. Then measured the diameter of the ring inhibited Helicobacter pylori.(4) Experimental research on healthy immune milk for infecting animals in prevention:18 two-grade Wistar rats were randomly divided into six groups:healthy immune milk group,antibody milk group,traditional Chinese herb milk group, Chinese medicine herbs decoction group,milk group and NaCl group. These rats were watered with SSI liquid(1010·L-1)separately seven times.All rats were killed and detected gastric antrum mucosa and cultivate Helicobacter pylori after four weeks.(5) Experimental research on healthy immune milk for infecting animals in treatment:18 two-grade Wistar rats were randomly divided into six groups:healthy immune milk group,antibody milk group,traditional Chinese herb milk group, Chinese medicine herbs decoction group,milk group and NaCl group. These rats were watered with SSI liquid(1010·L-1)separately seven times.All rats were killed and detected gastric antrum mucosa and cultivate Helicobacter pylori after four weeks.(6) The clinical observasion of healthy immune milk: 64 patients with HpAG were randomly divided into 4 groups: healthy immune milk,antibody milk,traditional Chinese herb milk and milk, every control group owned 16 cases, of which the treatment group was treated with Bismuth potassium citrate , Metronidazole, Amoxicillin , and healthy immune milk,antibody milk,traditional Chinese milk,last for two weeks. Examined Hp with 13C experiment after four weeks. Afterwards the patients who didn't eat anything drink the former milk before sleeping for eight weeks.Then examined Hp with 13C experiment.After treatment, detected the eradication rate of Helicobacter pylori, the rate of the second time reflection, clinical symptom and side effect.Results: (1) The result on relative antibody of milk: the milk wasn't measured antibody that inhibit Helicobacter pylori before immune. After the tenth day of the first immune, the milk from three cows all has low effective antibody, four times later the titer got to 1:2048 in direct agglutination test and 1:32 in double agar diffusion and lasted for one month.(2) The conclusion of the traditional Chinese composition: Others form the line of precipitate with corresponding traditional Chinese decoction in milk appeared in different degree except gongying with double laminar analysis diffusion.(3) Experimental research on inhibitive action on Heli- cobacter pylori in vitro:the former four groups all have obviously restrained effects on Helicobacter pylori,in which the group contains antibody and milk was better than the group only contain antibody or milk.(4) Experimental research on healthy immune milk for infecting animals of Helicobacter pylori in prevention:Two indexes that contain healthy immune milk and antibody showed negative;Two indexes contain traditional Chinese milk and additional Chinese decoction showed positive weakly;Two indexes contain milk and NaCl showed positive strongly.(5) Experimental research on healthy immune milk for infecting animals of Helicobacter pylori in treatment:Two indexes that contain healthy immune milk and antibody milk showed positive weakly;Two indexes contain traditional Chinese milk and additional Chinese decoction showed positive.Two indexes contain milk and NaCl showed positive strongly.(6) Clinical observation:The auxiliary efficiency of healthy immune milk for traditional Chinese symptoms: The healthy immune group and traditional Chinese milk group were better than the control group in the total availability ratio. There were distinct differences (p<0.05). Antibody milk group was lower than the former two groups but better than the control group. There were not distinct differences (p > 0.05 ).The auxiliary efficiency of Healthy immune milk for herding Hp: The healthy immune milk and antibody group with chemical medicine was better in herding Hp. Traditional Chinese milk group was better than the control group but there were not distinct differences compared to the control group(p > 0.05).The effect of the healthy immune milk for preventing the second reflection: The healthy immune milk and antibody group all were not discovered to be infected once more, which have distinct differences compared with the control group (p<0.05). There are not many patients who were infected in the herb group but there were not distinct differences compared to the control group(p > 0.05).The condition that the auxiliary treatment reduced side effect: The healthy immune milk or traditional Chinese milk with chemical medicine all could reduce the side effect ,of witch there were distinct differences with the control group (p<0.05). The effect of only antibody milk on reducing side effect was better than the control group but there were not distinct differences (p > 0.05).Conclusion: The milk has high titer antibody and last for thirty days through four times of immuning cows and the high titer was keep only by immuning every month.Serum and milk were found the corresponding composition of the traditional Chinese medicine by feeding traditional Chinese Compound to cows twelve hours later.The healthy immune milk had obvious effect on restraining Helicobacter pylori in vitro;Antibody and the traditional Chinese medicine may had cooperative effects on restraining Helicobacter pylori. It had treatment effect on animal inflected with Hp to some extent but it was difficult to eradicate;It had good prevention effect on animal to be inflected with Hp.The healthy immune milk with chemical medicine had remarkable effect on improving traditional Chinese symptoms; It reduced side effect of chemical medicine; It may have the auxiliary effect on eradicating Hp; It had reliable effect on preventing infection again after Hp was eradicated.
