Effect Of Replenishing-spleen Principle Of Changes Of The Structure Of Erythrocyte Membrane,ATPase Activities And The Ultrastructure Of Hippocampi In Old Spleen-deficiency Rat

The relationships among the spleen deficiency, senility, free radical damage, changes of the structure of cell membrane and functions, changes of activity of ATP-ase and apoptosis are studied with the method of molecular biology, in order to find out the possible mechanism of treating senility with methods of replenishing spleen. Methods:The young and senile rat models with spleen Qi, spleen Yang and spleen Yin deficiency syndrome are set up successfully by applying raulreiply factors such as abnormal diet, overstrain and damage from abuse administration drugs, which are treat with replenishing spleen methods. Observing markers include: (1) content of lactoperoxidase (LPO), superoxide dismutase (SOD) and lipofusc(LPF) in in plasm; (2) The content of phospholipid(PL), cholesterol(Ch), phosphatidylcholine(PC), phosphatidylethanolamine(PE), sphingomyelin(SM) in erythrocyte membrane. (3)The active of Na+-K+-ATPase > Ca++-Mg++-ATPase, Ca++-ATPase in erythrocyte membrane". (4) The ultrastructure of Hippocampi. Results:Compared to the control group: the content of LPO increases, the active of SOD decreases, the content of LPF increases. The content of Ch and SM increase (P<0.05), but there are no significant difference in PC and PE. The ultrastructure ofHippocampi suggests the cell apoptosis and senile changes occur. Chinese herbs of replenishing spleen can change the markers in different levels, such as increasing the content of PC, decreasing the SM and SM/PC, increasing the active of Na+-K+-ATPase, Ca++-Mg++-ATPase, Ca++-ATPase in cell membrane, ameliorating the ultrastructure of Hippocampi. Conclusions:(l)Spleen and stomach deficiency are the important causes of senility, and Chinese herbs of replenishing spleen can ameliorating the marker of senility and postpone the occurrence of senility effectively.(2) The possible mechanism of treating senility with methods of replenishing spleen include: (1) the function of anti-peroxidation. (2) protecting and repairing the cell membrane, so as to postpone the senility of cells,(3)increasing the active of Na+-K+-ATPase,Ca+-Mg+-ATPase, Ca++-ATPase in cell membrane. ?regulating the macrobiosis gene expression, slowing down the process of apoptosis, ameliorating the ultrastructure of Hippocampi.
