Study On Treatment Mechanism Of Diabetic Nephropathy With Tang-Shen-Kang

In this project, we made animal model of diabetes with frequent STZ injection, feeding high fatty feedingstaff, then observed indexes of blood sugar, renal function, kidney hypertrophy, glomerular extracellular matrix, cell factor on the end of the twelfth weeks, the sixteenth week and the twentieth week with advanced technologies such as micropicture analysis system, immune-tissue chemistry, ELISA, optical microscope and electronic microscope. With comparing the influence of Tang-Shen-Kang on indexes in different stages and impairment of kidney, we discussed treatment mechanism of DN with Tang-Shen-Kang, the results: (1) Rats with diabetes has glomerular mesangium hyperplasia, basement membrance hyperplasia accompanying with lipid metabolic disorder, peroxidation, all the indexes begin to increase from the twelfth week and reach peak till the end of twentieth week. (2) Tang-Shen-Kang has effects of reducing blood sugar, urinary albumin amount (24h), urea nitrogen, creatinine clearance rate, glycosylation hemoglobin content, the rats with big dosage are obvious (p<0.01), then middle dosage (p<0.05), the effects are as similar as control group with western medicine (p>0.05). (3) Tang-Shen-Kang has effect of preventing hypertrophic kidney of rats with DN in different stages. The following data of rats in treatment group with big and middle dosage of Tang-Shen-Kang and in control group with west medicine , kidney-weight/ bogy-weight, glomerular diameter and volume, reduced obviously, especially for rats with big dosage (p<0.01), data of rats with small dosage also have lowing tendency, but no statistic significance (p>0.05); (4) Tang-Shen-Kang with big dosage and western medicine can inhibit glomerular mesangium hyperplasia, basement membrance hyperplasia, both of them have effect of reducing fibronection, laminln, and collagen IV;(5) The indexes, 6-Keto-PGFl a /TXB2 and ET-1 in blood plasma, MDA content in kidney tissue, TNF- a ,IL-6 in blood serum began to increase from the end of twelfth and reach peak till the end of the twentieth, the result showed that DN can cause to high GFR, hypertrophy, glomerular cirrhosis, severe kidney impairment by different mechanism. Tang-Shen-Kang can treat and prevent DN by regulating the above indexes, which also indicated a certain relation between dosage and effective period.
