| Purpose:To confirm the effect of Tang Shen Kang decoction serum on the apoptosis and NLRP3 inflammsome of glomerulopodocytes induced by high glucose in vitro.This article is preliminarily discussed the mechanism of Tang Shen Kang decoction in treatment of early diabetic nephropathy so that providing theoretical basis for guiding clinical practice.Methods:1.MTT method was used to detect the effect of high glucose on podocyte apoptosis and the effect of Tang Shen Kang decoction serum on the apoptosis of podocytes induced by high glucose.2.Analysis of the rate of podocyte apoptosis.3.The concentration of IL-1? were detected by ELISA method.4.Detection of the protein expression of NLRP3,ASC and Caspase-1 activity in the supernatant of podocytes by Western Blot.Results:1.High glucose can induce podocyte apoptosis and has a certain correlation with time.2.The serum of Tang Shen Kang Yin decoction serum significantly reduced the apoptosis of podocyte induced by high glucose.3.The analysis of cell cycle by flow cytometry showed that the apoptosis rate of podocytes could be significantly inhibited by Tang Shen Kang decoction serum.4.The concentration of IL-1 beta produced by podocyte in high glucose group was significantly higher than that in normal control group(P<0.05),and the concentration of IL-1 beta in high glucose and decoction serum group was significantly lower than that in high glucose group(P<0.05).5.The protein expression of NLRP3,ASC and Caspase-1 in the high glucose group was significantly higher than that in the normal control group(P<0.05),and the protein expression in the high glucose and decoction serum groups was significantly lower than that of the high glucose group(P<0.05).Conclusion:1.Tang Shen Kang decoction serum can inhibit the apoptosis of podocytes.2.High glucose can induce the activation of NLRP3 inflammasome.3.Tang Shen Kang decoction maybe slow the progress of early DN by inhibiting the expression of NLRP3 inflammsome,which reduce IL-1?released. |
