Purpose: To explor the the protective effect of the eliminating phlegm and removing blood stasis on apoptosis by inner ear pathological of the early diabetic rat induced by Alloxan,and preliminarily probe into the possible inner ear pathological and hearing loss induced by machnism of diabetes and effect of The eliminating phlegm and removing blood stasis on this disease.Method: Induce diabetic hearing loss rats models by introperitoneal injection of middle dosage Alloxan.Treat the diabetic hearing loss rats with high and low dosage of The eliminating phlegm and removing blood stasis liquid respectively through tube feeding and at the same time set up the control group, the diabetic model group-, west medicine group(glibeclamide) and complex west medicine group(glibeclamide and duxil),some technology and methods are acquired,include of cochlear isolation methods, cell culture methods, immunohistochemistry, auditory brainstem respose(ABR), terminal-deoxynuci eotidy transferase mediated d-UTP nick-end labeling(TUNEL)technology, in situ hybridization laser scanning confocal microscope and electron microscopic. To examine the effects of The eliminating phlegm and removing blood stasis on anti-diabetic hearing loss, the rats were induced diabetic hearing loss by Alloxan.The mechanism of treatment of anti-cochlear apoptosis to The eliminating phlegm and removing blood stasis were investigated from cytokines and signal transmission.Results: The experimental rats are apparently improved in following parameters general condition are decrease blood glucose concentration,hemorheology lipid metabolism.The eliminating phlegm and removing blood stasis can decrease the ABR threshold of two TCM group .It can reduce the free calcium concentration in the outer hair cells of experimental rats cochlea.There was a lot of NOS c-Jun Fas protein in hair cell ganglion spiral cell and stria vascularis cell,The difference of NOS c-Jun Fas between TCM group and control group was significant(P<0.05).There was a lot of bFGFmRNA receptors gene in supporting cells of rats cochlea,but there wasn' t in normal group. Apoptosis was happened in hair cell, ganglion spiral cell and stria vascularis cell in cochlea of diabetic rats after being tested with TUNEL technology. We can observe ganglion spiral cell in cochlea of diabetic rats by eiectron microscopic. But there was no apoptosis in control group.Conclusion: The cochlea and central hervous system damage might be relate to hypercoagulability of blood in diabetes. The eliminating phlegm and removing blood stasis can efficientiy prevent the diabetic microangiopathy the microcirculatory disorders in the diabetic rats,and improve the thick wall of microvessel pathological progression. The eliminating phlegm and removing blood stasis can inhibit the diabetic-induced apoptosis of outer hair cell, ganglion spiral cell etc, in the cochlear of rats, improve the auditory function.
