Euonymus alatus is the dried winged branch and its attachments of Euonymus alatus(Thunb.)Sieb. that is in Celatraceae family. The drug is bitter, acrid in flavor and cold in nature.It has the effect of activating blood to resolve stagnation.The experiment selected H2O, 50% EtOH, 95% EtOH as elutes, using macroreticular resin column chromatography to sift active position of antimyocardial ischemia.Combining with the detection methods of screening test, biochemistry and immunohistochemistry, the article explored their imfluence about cell apoptosis and genetic expression.It came to a conclusion that 50% elute and 95%elute appeared obvious protection to myocardial ischemia, reduced incidence of myocardial cell apoptosis, induced the expression of apoptosis suppressor gene bcl-2 and inhibiting apoptosis stimutus gene bax.Two elutes had no singnificant difference. Combining with the test result of rate of recovery, the article selected 50% elute as main active position to protect from myocardial ischemia damage.With conducting systemic studies, chemical constituents were isolated and identified.The selected active position proceeded various chromatogra-phic methods. 25 chemical constituents were isolated and identified . The 50%elute was partitioned with EtOAc, BuOH and H2O successively. The EtOAc fraction was chromatographed on silica gel column, Sephadex LH-20 column chromatography and HPLC to give compounds 1-17, and 19-24, while the H2O fraction was separated Polyamide column chromatography and silica gel column to afford compounds 18,25.On the basis of analysis of UV, NMR, MS data, measurement of physico-chemical characters and chemical method,the structures of 18 compounds were identified as 4 organic acid, 2 alkaloid, 5 lignin, 3 flavone, 4 glycoside. Among them, 14 were first isolated from this plant, they are 5-methoxy-2-furaldehyde (l),alanine-proline lauroxixanes (2), 3, 5-dimethoxy-4-hydroxybenz-aldehyde (3), benzoic acid (4), episyringaresinol (7), syringaresinol (8), diapinoresinol mono methy erther(9), pinoresinol mono methy erther( 10),ciwujiatone( 11 ),eugenyl-o- -D-glucopyranoside( 12), ade-nosine(13), phenethyl alcohol-8-O-p-D-gentobioside(14), 3-O-a-D-glucopyranoside-3,4-dihydroxyhypnone(15).Among them, compounds 1, 2, 3, 7, 9,11, 12, 14, 15 were isolated from this genus for the first time. The known compounds are Hyperin(16), (+)-Catechin(17), hesperidin(18), p-hydroxybenzoic acid(6), 3, 5-dimethoxy-4-hydroxy benzoic acid(5).The results supplied the researching base to study chemical constituents, biological activities, pharmacy-material, and the development of Euonymus alatus.
