Objective:To investigate the chemical constituents from Euonymus alatus (Thunb.) Sieb and their hepatoprotective activity.Method:Compounds were islolated by repeated silica gel, Rp C18and sephade LP-20column chromatographies. Their stractures were elucidated by spectroscopic methods. The protective effect of the isolated compouds on Hep G2cell induced by tert-butyl hydroperoxide was evaluated.Result:Six compounds (1-6) from the Ethyl acetate extract layer, and one compound (7) from the Petroleum ether extract layer of E. alatus alcohol extract. Their structures were identified as catechin (1), quercetin (2), hyperoside (3), kaempferol (4), geniposide (5), triptolide (6) and methyl salicylate (7). Four compounds (1-4) showed significant hepatoprotective activity with EC50values of40.24μM,29:56μM,23.82μM and5.41μM, respectively, on tert-butyl hydroperoxide induced cytotoxicity in Hep G2cell line.Conclusion:Four flavonoids from E. alatus showed the hepatoprotective effects on tert-butyl hydroperoxide induced cytotoxicity in Hep G2cell.
