The Pancreatic Carcinoma has the worst Prognosis in the digestive tract. Its Five year survival rate is less 5 percent. The reasons of worst prognosis are the invasion and metastasis of the tumor. Recent researches show the invasion and metastasis of malignant tumor is a multi-factors, multi-steps, multi-phases complicated process. And the change of the tumor's adhesion plays an important role in the process of invasion and metastasis.Huoxue Huayu therapy is a main way of treating malignant tumor with Chinese medicine. In the Chinese medicine views, the Yuxue Neizu is an important reason in the carcinogenesis. Many clinic and research studies make sure the immediate relations between the carcinogenesis and metastasis of tumor with the Xueyu symptom in patients. Therefore, Huoxue Huayu therapy accords with the principle of Bianzheng Lunzhi in the Chinese medicine. Ligusticum Chuanxiong Hort, a typical Huoxue Huayu Herbs, can play a role of anti-tumor effects by immediately restraining tumor, reducing the toxic and increasing the effect of chemotherapy, can play a role of anti-metastasis effects by anti-cruor andreducing the platelet adhesion reaction. But there are many puzzles about the influence of Ligusticum Chuanxiong Horton invasion and metastasis of tumor, and there are many disputes on its effects of adhesion molecular and angiogenesis. By the way, we studied the effects of Ligusticum Chuanxiong Hort on the pancreatic carcinoma growth, invasion and metastasis by in vivo and in vitro.To simulate the biologic characters of pancreatic carcinoma in vivo, we first built an orthotropic pancreatic model in nude rats. In the base of cytological study in vitro, we study the effects of Ligusticum Chuanxiong Hort on the growth, invasion and metastasis of pancreatic carcinoma in vivo by animal model, which set up the foundation of the study on the Huoxue Huayu treatment malignant carcinoma.Part I : Experimental study on effect of the Ligusticum Chuanxiong Hort on proliferation and adhesion of pancreatic cancer cell HS 766T in vitroAim: To study the effect of the Ligusticum Chuanxiong Hort on proliferation and adhesion of pancreatic cancer cell HS 766T in vitro, and determine the dose of Ligusticum Chuanxiong Hort in part II.Methods: We built the pancreatic cancer cell group HS 766T and human endothelium cell in the umbilical vein by the way of cell culture in vitro. The effect of Ligusticum Chuanxiong Hort on the proliferation of HS 766T cells was examined by MTT method. The changes of the cell cycle and apoptosis of HS 766T cells dealing with Ligusticum Chuanxiong Hort were determined by PI dyeing and flow cytometry. We examined the effect of Ligusticum Chuanxiong Hort on the adhesionbetween the laminin and the HS 766T cells by the way of CCK-8 method, determined the effect of Ligusticum Chuanxiong Hort on the adhesion between the human endothelium cells and the HS 766T cells.Results: Ligusticum Chuanxiong Hort can restrain the proliferation of pancreatic cancer cell HS 766T in vitro as direct relation of time and dosage. Ligusticum Chuanxiong Hort can promote the apoptosis of the HS 766T cells by blocking the G0/G1 phase cells, restraining cellular proliferation and increasing the percent of synthesize phase cells. Ligusticum Chuanxiong Hort can restrain the invasion of HS 766T cells in vitro. Ligusticum Chuanxiong Hort can restrain the adhesion ability of the HS 766T cells with the laminin and the endothelium cells as relation of time and dosage.Conclusion: Ligusticum Chuanxiong Hort can restrain the proliferation, invasion and adhesion ability of pancreatic cancer cell HS 766T in vitro.
