| Diabetes mellitus(DM) is a kind of endocrinal metabolic disease caused by the decreased in the synthesis and excretion of insulin, or the insulin receptor and post-receptor defects.Mini-pigs were qualified for small body type, stability of heritage, resemblance in organ function and tissue structure with human, easily controlled experiment condition. The mini-pig resource is abundance in China. Research showed that there were 24 blood physiological index of Bama mini-pigs similar with human, which indicated that physiologic function of Bama minipigs had similarity with human. The value of blood and urine of SPF mini-pigs had tiny alternation range. The blood constituent in SPF mouse had conformity change tendency with SPF mini-pig. STZ selectively destroyed insular β cell of some genera animals and induced the Diabetes mellitus, the stomach intestine dyskinesis animals model might created by peritoneal injection of Alloxan(40mg/Kg) while feeding with high glucose stoyer. So these two kinds of animal model are widely used in DM research. Cell apoptosis had a very important position in the development of DM compalication. With the status of high blood sugar, the expression of anti- apoptosis, Bcl-2 and Bcl-x was decrease. On the contrast, the expression of promoting apoptosis gene-Bax was increase, which casued the increase number of apoptosis cell. The research aim was to investigate the key method to prevent and cure DM complication through the research of adjustment of apoptosis.Traditional Chinese Medicine TCM provided many experiences on the treatment of stomach dyskinesis of dibetes. Its basic pathogenesis was spleen malfunction of transportation and transformation,and the causative factors were blood stasis, turbid phlegm, Qi stagnation, and damp obstruction, etc. It pointed that the deficiency of kidney essence and kidney yang, as well as the deficiency of spleen yang and spleen qi might lead to the spleen malfunctionof transportai ion and transformation. Qi depression and stagnation of stomach was an important pathogeneses of stomach dyskenesis of diabetes. Patients with stomach dyskenesis of diabetes expressed some digestive tract symptoms including distention .vomiting , belching , stomach reflux , nausea , phlegm , and tardy stomach . The same in other patients who did not gain stomach dyskenesis of diabetes. Even with the same symptoms, the level was different . TCM promoted the theory of invigorating spleen, regulating qi and activating blood circulation to dissipate blood stasis to deal with stomach dyskenesis of diabetes. Diabetic gastroparesis belong to root-deficient and branch-sufficient syndrome. The Chinese composition piweikang was an experienced prescription which has significant curing effect in clinical practice.In order to discuss the procedure and method to establish the DM animal model of minipigs, to reveal the nosogenesis of diabetic complication and to reveal the drug action mechanism of piweikang, The research was launched.The aim was to research the feasibility to establish DM animal model with Bama minipigs through the observation on physiological and biochemical index, alimentary canal ultra-micro tissue architecture, and to establish the surgeon procedure for implanting electrodes into stomache and the detection method of gastric electrical activity, and to duplicate the DM animal model in minipigs and to observe the change of pathology in DM model rats stomach tissue and the expression change of Bax^ Bcl-2 to investegate the mechanism of Traditional Chinese Medicine composition piweikang on the prevention and cure of Diabetes MellitusThe results showed that growth and development was halt or torpidity at 12 months age in minipigs. With small body type, stability of heritage, resemblance in organ function and tissue structure with human,gentle temptation, and easily controlled, minipigs were useful experimental animals.Special electrodes were implanted into stomache of mini-pigs under combined anesthesia. During the operation period,there was no abnormal reaction,the success operation rate was 95% and the animal had excellent postoperative recovery. The results indicated that as a new experimental animal on the research of gastric electrical activity , minipigs was safe and easily handling.The normal gastric electrical act ivity of Bama minipigs was first reported. |
