After binding to each other, the axis of this recepter and ligend will accelerate the inflammation and immunologic reaction by adjusting the synthesis of many substances, and then induce the atherosclerosis occurrence and progress. The blockade of CD40/CD40L receptor and ligend axis may contribute to the control the occurrence and progress of atherosclerosic plaque.RNA interference is a new method of gene silencing with the characters of specificity,effectivity and conveniece. It was selected that the mRNA target with the best inhibiting effect.The CD40L siRNA lentivirus was established successfully based on the selected target. The outcome are lower sCD40L and CD40LmRNA; less CD40L and MMP-9 in local vessel with immunohistochemistry than the control groups; Compared with the model lentivirus control group, there are less atherosclerosic plaque in siRNA group, and the plaque depth and area are decreased.
