Chronic renal failure (CRF) is a refractory diease known as a progressive and irreversible process. Renal replacement therapy is a way to cope with CRF, but it is very costly and impacted by somewhat immunal rejection, thus not applicable extensively. Therefore, measures to delay CRF exacerbation are essentially important. Neuclear factor kappa B (NF-κB) was proved playing important role in CRF progression, so that expression of NF-κB under medication is essential to assess the effects of the medicine being used.In this study, the effects of Chongcaoshencha Prescription (CCSCP) were examined in the rat with 5/6 subtotal nephrectomy (SNx) controlled by Fosinopril ( Monopril ). NF-κB, AT1 and Nestin were examined by immunohistochemical semi-quantitative technique on the remaining renal tissues. ON the basis of experiments above, the pathogenesis and mechanisms of CRF were discussed. In addition, a clinical observation of 60 patients with chronic kidney disease medicated with CCSCP at different phases was also carried out. The medication time with CCSCP was then further clarified.Result indicated:1. CCSCP was proved an effective formula in treating CRF.2. 5/6 Nx was an ideal animal model of CRF of glomerulopathy.3. CCSCP enchanced re noprotective in 5/6 Nx, and the mechanism may included suppressing glomerular hypotrophy, inhabiting the expression of AT1 and Nestin, and reducing proteinuria excretion.4. CCSCP was effective in inhabiting the activity of NF-κB, reducing the impair resulted from a series of immunoinflammation induced by NF-κB, and delayong the progression of CRF.5. Overall assessment of CCSCP in treating CRF based on the mechanism of multiple targets and loops was better than monopril.6. Both experimental research and clinical observation proved that CCSCP was functional to postpone the progression of CRF, and early medication resulted in better renoprotecing.7. The theory of ascending and descending Qi in recognition of CRF is scientific and proper.In conclusion, NF-κB plays a very important role in the progress of CRF. CCSCP is an effective formula to postpone the progression of and prevent CRF by regulating the expression of NF-κB and related cellular factors. CCSCP is worthy of further study.
