Objective:To evaluate the intervention of Huanglian Wendan Decoction on various metabolic disorders in metabolic syndrome (MS) rats comprehen-sively; To research its influence on NF-κB,PPARa and Adipocytokines; To explore its molecular mechanisms on MS prevention and controlMethod:we established the MS model by feeding the SD rats high glucose, high fat and high salt diet for 20 weeks,then chose 37 rats who reached the MS model standards. Those MS rats were into MS Model Group (MSG), Fenofibrate Group (FG), HWD High Dosage Group(HDG) and HWD Low Dosage Group (LDG).After the corresponding drug therapy for four weeks, we tested indexes such as weight, abdominal perimeter, blood pressure, FBG, FINS, blood-fat, etc, and Figured out the HOMA-IR to evaluate animal models and drug interactions. Before and after drugs treatment we detected the serum level of TNF-α,Leptin and IL-6 with Radioimmunity Assay, the expression of VCAM-1 and NF-κB in aortas of rats with Immunohistochemis-try Assay,the serum level of FFA by Colorimetric method, and the expression of PPARamRNA with RT-PCR Assay.Result:The weight and blood pressure of the rats in experimental groups with high glucose, high fat and high salinity diet grew obviously than those in normal groups after 8 weeks (P<0.05). Till the end of 20 weeks, the serum insulin, TG, TC and LDL-C of the rats in experimental groups all increased evidently (P<0.05), and their HDL-C reduced inappreciably (P>0.05).Among the MS rats, the data of the weight, abdominal perimeter, blood pressure, FBG, FINS, blood-fat, HOMA-IR, TG, TC and LDL-C all increased and the level of HDL-C reduced evidently (P<0.05).At the end of pharmacological intervention, the data of weight. Lee's index. VFM, weight ratio of visceral fat, blood glucose, serum insulin, HOMA-IR. TG. TC and LDL-C of HDG and FG had a greater drop in yield than those of MSG (P<0.05). Only the blood pressure of HDG decreased obviously (P<0.05). Only Lee's index and TC of LDG had a greater drop in yield than that of MSG (P<0.05). There is no significant difference among the groups about the cardiac rate and body length of the rats (P>0.05). HDL-C of FG had significant difference between those data of MSG (P<0.05). The serum TNF-α, IL-6, Leptin, FFA, and the expression of VCAM-1 and NF-κB in aortas of MSG rats all increased notably (P<0.01). After 4 weeks of pharmacological intervention, all the above indices of HDG and FG notably decreased (P<0.01). Only the vessel FFA, and the expression of VCAM-1 and NF-κB of LDG decreased notably (P<0.05). The expression of PPAR-amRNA of HDG and FG remarkably decreased (P<0.05).Conclusion:1. The MS rats model can be successfully established by raising the SD rats with high glucose, high fat and high salinity feed for long time. And the model has basic MS feature.2. Huanglian Wendan Decoction can decrease the blood pressure, blood glucose and weight, adjust blood-fat, relieve Insulin Resistance, and improve comprehensively various metabolism disorders in MS rats. It calls attention that the modification in HWD could become one of the effective medication used to prevent and control MS.3. Huanglian Wendan Decoction can inhibit NF-κB being activated in aortas of rats with MS and decrease the expression of adipocytokines VCAM-1, TNF-αand IL-6 in various degree, thus alleviate inflammatory reaction.4. Huanglian Wendan Decoction can decrease adipocytokines FFA and Leptin level, mend Insulin Resistance and dyslipidemia by downward modu-lating the expression of liver PPARamRNA of MS rats5. Downward modulating the expression of liver PPARamRNA, inhibit-ing NF-κB being activated in aortas, decreasing adipocytokines Leptin, FFA, TNF-a. IL-6 and VCAM-llevel, alleviating inflammatory reaction, and improving Insulin Resistance, all above could be one of the mechanism of Huanglian Wendan Decoction which could be used to prevent and control MS and early period of vascular lesion.
