Objective:To evaluate the treatment of Huanglian Wendan Decoction on the disorders of metabolic syndrome in rats and study its influence on PI3K signal pathway of skeletal muscle. To explore the molecular mechanisms on the PI3K pathway and the relationship between adipocytokines and this signal pathway.Method:The MS model was completed by dieting the SD rats with high glucose, high fat and high salt diet for20weeks, then treated the rats with streptozocin(STZ,30mg/kg)by intraperitoneal injection. Those MS rats were divided into MS model group, western medicine group, high dosage group, middle dosage group and low dosage group by random and each group was lavaged the corresponding drugs treatment for four weeks. The indexes such as weight, abdominal perimeter, blood pressure, FBG, FINS, lipid, etc, were detected and figuring out the HOMA-IR, Lee's to evaluate animal models and drug interactions. Before and after drug treatment the level of FFA, TNF-a, LEP, the expression of IRS-1, GLUT-4and PI3K of skeletal muscle in those rats were detected.Result:1.Compared with the control group, the rats feeding high glucose, high fat, high salt diet and treated with STZ, the level of weight, abdominal perimeter, blood pressure, FBG, FINS, TC and TG was increased and it has statistical significance (P<0.05)2.After treated with the drug, compared with the control group, the weight, blood pressure, Lee's index, visceral fat weight, the proportion of visceral fat and weight, FBG, FINS, HOMA-IR, TG, and TC has statistical significance (P<0.05). But the indexes of other groups have no difference.3.After treated with the drugs, compared with the control group, the level of FFA, TNF-α, LEP has statistical significance (P<0.05), but there is no statistical difference in other groups of these indexes.4.After treated with the drugs, compared with the control group, the expression of IRS-1mRNA in skeletal in high dosage group is obviously higher and has statistical significance (P<0.05)5.After treated with the drugs, compared with the control group, the protein expression level of IRS-1, PI3K, GLUT-4was significantly higher and has statistical significance (P<0.05)6.According to the relative research, there is associativity between FFA, LEP, TNF-a and IRS-1, which is negative correlation, especially obvious with IRS-1and LEP (r=-0.641, P<0.01)Conclusions:1.Huanglian Wendan Decoction can decrease the level of MS rat's weight, blood pressure, blood glucose, FINS and relieve Insulin Resistance, regulate the lipid level and improve the various metabolism disorders in MS rats, especially in high dosage group.2.Huanglian Wendan Decoction can reduce the adipocytokines in different extent such as FFA, LEP, TNF-a and then inhibited the inflammation response. This effection may be relative with reducing the weight, blood glucose, lipid level and Insulin Resistance.3.Huanglian Wendan Decoction upregulated the IRS-1mRNA in sleletal muscle of MS rats, and actived the IRS-1-PI3K-GLUT-4signal pathway, increased the level of absorbing and using glucose, improved the Insulin Resistance.4.Huanglian Wendan Decoction upregulated the protein of IRS-1, PI3K, GLUT-4in skeletal muscel of MS rats, and then improved the insulin signal pathway to ameliorate the glucose metabolic and IR, which may be one of the mechanisms to prevent MS.
