Part I The expression and clinical significance of ATDC in the large intestineObjective:TO investigate the impact of ataxia telangiectasia group D complement gene (ATDC) in the development of colon cancer and their clinical significance.Methods:The specimens were divided into colorectal cancer group and normal colorectal tissues(control group).Immunohistochemical methods was used to detect expression and localization of ATDC protein. Western blot and quantitative RT-PCR were used to detect mRNA and protein expression of ATDC.The relationship between ATDC expression and clinicopathological parameters by PASW Statistics 18.Results:The ATDC protein expression in normal tissue and cancer tissue were statistically significantly different(P<0.05).The ATDC expression has no relationship with gender,age and dukes stage except lymph node metastasis and the degree of differentiation.Conclusion:The high expression levels of ATDC in colorectal carcinoma,suggest that genes involved in the process of tumor development.Detection and suppression of ATDC expression have great significance for the diagnosis and treatment of tumors;the realation of ATDC and the clinicopathological parameters of colorectal cancer,suggest that ATDC at high levels of expression may have some clinical value for assessment of colorectal cancer treatment and prognosis. Partâ…¡Effect of silence of ATDC expression for biological characteristics in colorectal cancer cellsObjective:To explore the silence of ATDC expression affect biological characteristics of colorectal cancer cells.Methods:ATDC-siRNA was transfected into SW-620.Real time PCR and Westermblot were used to detected mRNA and protein respectively.Cell cycle and apotosis were investigated by FCM. With the use of transwell invasion assay,the invasive property of cancer cell was detected.Result:Expression of mRNA and protein were downregulated in siRNA-group,compared with control-group.Compared to bland-group and negative controlsiRNA-group,the siRNA-group most cells stag at G0/G1 phase and less at G2/M phase.Transwell invasion assay suggested downregulation of invasion and metastasis in cancer cells.Conclusion:Sileneing of ATDC lead to cancer cell cycle arrest and attenuate malignant phenotype...
