Female breast is not only a nursing organ but also an organ of esthetics and sex. Hypomastia not only affects women's feeding fimction, but also has very adverse influence in the development of their healthy psychological status, good social adaptability and family relation. With the development of social economy, civilization and change of traditional value, female pursues high quality of life. Therefore ill-developed breast becomes an irnpoitant element which affects female's life. However the existing treatments including operation and hormone therapy, with its obvious defects, can not meet the social demand. In the last few years, great achievement has been made in using kidney reinforcing and blood circulation activating Chinese medicine (K&B) to restore the balance of reproductive endocrine system and promote the development of sexual hormone's target organ-the uterus. But little research has been carried out to probe the effect and mechanism of K&B in the development of mammaiy glands. In the theoretic investigation part of this research work, structural formation and influence factors of breast's form have been studied. It reveals that it is of more significance to study the status of estradiol reccptors(ER) and progesterone receptors (PR) of mammary glands than to simply analyse the serum hormone level in terms of investigating effect and mechanism of K&B. The approach of Radioligand Binding Assay of Receptor (RBA) is more suitable in conducting the above research at the pre-clinical level. In addition, relying on the results of previous modern combined Chinese and Western laboratory and clinical research, K&B's safety and protection to the mammary glands has been discussed. The laboratory research includes part I, II and m. The objective of part I is to study the effects and mechanism of K&B in mannnaiy glands development. Methods: Using the female immature rats as the experimental targets, the level of serum hormone, the amount and affinity of ER and PR as well as the DNA content and relative weight of the rats?maminaiy glands were measured in studying the effects of K&B at the pre-reccptor, receptor and post-receptor level. Results: K&B markedly enhanced the scrwn level of estradiol, progesterone and the growth hormone (GH) at pie-receptor level, notably increased the number and affinity of estradiol and progesterone receptors both in the cytoplasm and nuclei at the receptor level. At the level of post-receptor, it increased the DNA content and the relative weight of mammary glands significantly. Conclusions: The kidney reinforcing and blood circulation activating Chinese medicine are effective in promoting the development of mammary glands. On the basis of pait I, the study of part II and III focuses on studying the effects and mechanism of K&B on the form and structure of mammary glands of atrophy and hyperpalsia models in mature female rats. Methods: The models of mammary glands atrophy and hyperplasia established respectively by ovariectomy and injecting large doses of Diethylstilbestrol were used as experiment targets to probe the effects of K&B on the relative weight and structure of mammaiy glands as well as the amount and affinity of ER and PR. Results: I). K&B notably ameliorated the atrophy of mammary glands and remarkably increased the relative weight of mammary glands comparing with that of model rats and normal rats, adjusted the ratio of EnR/EcR and PnRIPcR towards normal level. 2).
