Warm Heart Particles Treatment Of Chronic Congestive Heart Failure The Role Of Mechanistic Studies

Objective:Evaluating the effect of Warming Yang of WenXing granule and the therapeutic effect of curing chronic congestive heart failure,and exploring the mechanism of it's treatment.Methods:Introducting the way of intraperitoneally injecting Hydrocortisone to establish mouse model of syndrome of deficiency of kidney-YANG,introducing the same way of using Doxorubicin hydrochloride and Hydrocortisone to establish rat model of chronic congestive heart failure(CCHF) that was the type of syndrome of deficiency of heart and kidney-YANk.After that,clearing it's therapeutic effect and mechanism through memorizing hemodynamics index with the BL-420E biological function experimental systems;observing the change of pathologic histology of rat's heart under light microscope;and determining the content of AngⅠ,AngⅡ,ALD,ANP,ET,TNF by Radio immunoassay.Results:1.WenXing granule has evident effect of Warming Yang.The experimental result shows that the low and middle dosage group's result of wanning Yang is better than that of high dosage group.2.WenXing granule can extremely strengthen±LV dp/dtmax and LVSP,and at the same time debase LVEDP,and prove that WenXing granule can obvious improve and rectify the change of the hemodynamics of model rat.3.WenXing granule can expand peripheral blood vessels,enhance CO to reduce Renin secretion and interrupt the activation of RAAS through strengthen±LV dp/dtmax and LVSP, on the other hand debase LVEDP,avoiding to occur the phenomenon of ventricle reconstitution.4.The mechanism of reducing the level of plasma ANP is to debase LVEDP to improve the preload of left ventricle.The mechanism of reducing the level of plasma ET is through which debasing LVEDP to improve the gore of lung circulation and strengthening±LV dp/dtmax and LVSP to increase the infusing volume of kidney blood.5.The mechanism of reducing the level of plasma TNF is to interrupt the activation of RAAS through strengthening±LV dp/dtmax and LVSP,with it to reduce the TNF-αproduction of macrophage and obstruct the oxidation induction of TNF-α.Conclusions:1.WenXing granule has evident effect of Warming Yang,and initially reveals the conclusion of NeiJing that of "junior fire supporting vital Qi,superheat consuming Qi"2.WenXing granule can extremely strengthen contractile function of left ventricle, improve diastolic function of left ventricle,debase preload,debase myocardial consumption of oxygen,versatilely retrieve the change of hemodynamics of model rat.3.The mechanism of WenXing granule to treat chronic congestive heart failure is involved with reducing the level of ANP,ET,TNF and regulating RAAS.Sum up,the main therapeutic mechanism of WenXing granule to CCHF is multi-angle and multi-arrangement.
