Chemical constituents in the methanol extract of WCA pill were studied tosearch for the active constituents in WCA pill, the main components in the methanolextract of Fructus Aurantii (FA) and the metabolites in rat biological samples afteroral administration of the methanol extract of FA were analyzed and the influence ofherbs on the pharmacokinetics of main components and pharmacology related tobowel movement were studied to elucidate the compatibility in the formulae. Thedetailed results were as follows:Fourty bioactive constituents in the methanol extract of WCA were identified anda reliable and accurate method for the simultaneous determination of twelvecomponents was developed.There were31constituents identified in the extract of FA including2alkaloids,10flavonoid glycosides and19ploymethoxylated flavones. According to the UVspectrum and MS fragment character of main components in the methanol extract ofFA,18parent constituents and11metabolites were tentatively identified in ratbiological samples.The pharmacokinetics of rhein was studied following oral administration of purerhein, Radix et Rhizoma Rhei methanol extract (RR) and Radix et Rhizoma Rhei andSemen Crotonis Pulveratum mathanol extract (RR-SC) with approximately the samedose. It was found that comparing with that in monomer, both AUC and Cmaxof rheinin RR and different proportion of RR-SC were significantly different. The resultsindicated that comparing with the monomer, the concentration of rhein in the plasmawas increased in the group of RR whereas the tendency of AUC and Cmaxof rhein inRR-SC presented quite discrepancy depending on the different proportion of RR-SC.Single and multiple oral administrations of the extracts of RR and FA-RR wereperformed to determine the concentrations of anthraquinones in rat plasma. Expect forphyscion, four anthrquinones including aloe-emodin, rhein, emodin and chrysophanolwere detected in rat plasma of single oral administration of the extracts of RR andFA-RR, while all of five anthrquinones were analyzed in the samples of multiple oraladministrations. The results indicated that the significant differences thepharmacokinetics of anthraquinones were detected not only in the compatibility oftwo herbs but in the single and multiple oral administrations. The pharmacokineticdifferences of two flavonoid glycosides were detected after oral administration of theextract of single herb and different compatibility of Fructus Aurantii, Radix etRhizoma Rhei and Semen Crotonis Pulveratum. Comparing to pharmacokineticparameters of the single herbs administration, there were no significant differenceafter the compatibility of Fructus Aurantii and Radix et Rhizoma Rhei. However, the absorption of naringin and neohesperidin was increased dramatically after thecompatibility of Fructus Aurantii and Semen Crotonis Pulveratum.The effect of Fructus Aurantii and Semen Crotonis Pulveratum on the smoothmuscle constractions was investigated in vitro using the isolated intestine moded ofdifferent parts. The extract of FA decreased the contractile amplitude of all part of theisolated intestine, while the extract of SC has a biphasic action contracting andrelaxing intestinal issue only on isolated jejunum. Comparing to the extract of FA, thecompatibility of two herbs concentration-dependently suppressed the amplitude ofmuscle contractions more strongly which induced by acetylcholine.The main components in root extract were identified by HPLC-ESI-MS/MS. Thechanges in the kind of secondary metabolites of licorice root after spaceflight werefirstly reported. Three acute inflammatory models were utilized to compare theanti-inflammatory activity of licorice pre and post spaceflight. The licorice extractshowed the significant anti-inflammation activity. And after spaceflight thepharmacological activity of licorice got higher than that of the ground control one.
