Cervical cancer is one of the malignant tumours that threaten Gynaecological Health seriously.At present chemotherapeutic chemosynthesis drugs are used to treat cervical cancer,at the same time,which produce harmful side-effects to the body.Screening and seeking more effective,less toxic crude drug is research focus.It is reported that active ingredients of bupleurum root,flavescent sophora root, rhubarb,phellodendron bark play a role in preventing and treating some cancers,treating cervical cancer with crude drug has been rarely reported in our country.The objective of the present study was to evaluate that water extract of Radix Bupleuri.Radix Sophorae Flavescentis,Radix et Rhizoma Rhei,Cortex Phellodendri act on Hela cell of human cervical cancer,to study mechanism of drug action,to provide a theoretical basis for developing new drug against cervical cancer.The object of the thesis was Hela cell of human cervical cancer developed in vitro,which was worked at different concentration of Radix Bupleuri,Radix Sophorae Flavescentis,Radix et Rhizoma Rhei,Cortex Phellodendri decoctions,whose morphology was observed by optical microscope.The effect of four Chinese herbs on growth of Hela cell was detected using a MTT approach,and DNA variation of Hela cell was analysed by agarose gel electrophoresis,action results mechanism of four Chinese herbs decoction on Hela cell was preliminary studied.It was detected by MTT method that four Chinese herbs decoction inhibited from growing of Hela cells significantly (P<0.01),which was dependent on both concentration and time. Thereinto, inhibitory effect of Radix Bupleuri decoction 48h post drug action was more obvious than that 24h post drug action, inhibition rate was 52.90% at the concentration of 80ml/L;inhibition rate of Radix Sophorae Flavescentis was 23.17%,the highest inhibition rate was 57.19%. Inhibition rate at the concentration of 80ml/L Radix et Rhizoma Rhei decoction 48h post action was up to 55.06%;inhibitory effect of Cortex Phellodendri decoction on Hela cells was obvious,and inhibition rate was between 8.43~62.85%.Morphological observation of Hela cell which was dealed with 5,10,20,40,80ml/L (four raw drugs concentration) was observed that cells were polygonal,abnormal,adherent tightly in the control group;while they were rounded,sparse and increased in Radix Bupleuri, group;and adhesive rate was low, smudge cells were increasing markedly 48h post-action at high concentration (80ml/L) in Radix et Rhizoma Rhei group;In Radix Sophorae Flavescentis, Cortex Phellodendri.cells were round and decreased in its size,karyopyknotic,most of cells falled off and suspended in culture,smudge cells increased with prolonging of action durationThe results of agarose gel electrophoresis of DNA indicated that DNA ladder occurred in Radix Sophorae Flavescentis decoction group at the concentration of 10ml/L or above 10ml/L;electrobands of DNA ladder occurred,when Cortex Phellodendri decoction acted on Hela cell at the concentration of 40ml/L,80ml/L;there were weaker electrobands of DNA ladder at the Radix et Rhizoma Rhei decoction concentration of 80ml/L 48h post action;DNA of Hela cells ruptured at the Radix Bupleuri decoction concentration of 40ml/L,80ml/L 48h post drug action,DNA ladder was unobvious.The trial results showed that Radix Sophorae Flavescentis decoction, Cortex Phellodendri decoction, Radix et Rhizoma Rhei decoction had an effect on inhibiting the growth of Hela cell and inducing apoptosis of Hela cells.Especially, Radix Sophorae Flavescentis and Cortex Phellodendri decoction had obvious an inhibition effect,the highest inhibition rate of which was up to over 55%,presenting typical DNA ladder bands by agarose gel electrophoresis,and speaking volumes for having an effect of Flavescentis and Cortex Phellodendri decoction on apoptosis of Hela cells,and whose inhibition effect had precedence over that of Radix et Rhizoma Rhei and Radix Bupleuri within the range of adoptive concentrations in this study.The results obtained from this study established the foundation for the further research in applying four Chinese herbs to anti-tumor and provided successful experiences for developing and utilizing four Chinese herbs.
