Chapter 1 Expressions of lysophosphatidic acid receptors in the development of epithelial ovarian carcinomaObjective To deeply investigate the expression of three lysophosphatidic acid receptor:LPA1,LPA2 and LPA3 mRNA and protein in epithelial ovarian cancer.Method 134 samples of epithelial ovarian tissues by surgical removal in Zhongshan People’s Hospital during Mar.2012 and Dec.2014 were chosen.The expressions of LPA protein in ovarian tissues were detected by IHC and LPA mRNA and protein expressions were detected by RT-PCR and western blot.The ROC was drawn to assess the applied value of LPA2 and LPA3 in the diagnosis of ovarian cancer.Results The positive expression of LPA protein leaded to apparent brown cell membrane and cytoplasm,the colorable range and intensity of epithelial ovarian carcinoma cells were larger and stronger than that of normal and benign ovarian tumor cells.Both the LPA2 mRNA and LPA3 mRNA in epithelial ovarian cancer group were higher than that of normal and benign control group.The AUC of LPA2 was 0.992(95%CI = 0.984-1.000,P<0.001),when cut-off value was 0.625,the diagnostic sensitivity(96.3%)and specificity(97.9%)reached to the highest level.The AUC of LPA3 was 0.999(95%CI = 0.997-1.000,P<0.001),when the cut-off was 0.565,the diagnostic sensitivity(98.5%)and specificity(97.9%)also reached to the highest level.Conclusion LPA2 and LPA3 probably participate in the occurrence and progression of epithelial ovarian carcinoma.Moreover,the application of LPA2 and LPA3 receptors in the diagnosis of epithelial ovarian carcinoma achieves high accuracy.Chapter 2 Analysis of predictive value of Tim-1/2/3 mRNA/protein expression in development and relapse of primary epithelial ovarian carcinomaObjective To explore the relationship between Tim-1/2/3 and relapse of ovarian carcinoma and its predictive value.Method A total of 134 epithelial ovarian carcinoma patients,who have been treated in Zhongshan People’s Hospital from March 2012 to December 2014 were selected.We use qRT-PCR to test expression of Tim-1/2/3 mRNA.We use western blot to test expression of Tim-1/2/3 protein.We followed up tightly after initial treatment,grouping by disease condition of tumor development and relapse.We use SPSS 21.0 to the the data analysis.We use(x± s),student t test to deal with measurement data.We use percentage and χ2 to deal with enumeration data.P is bilateral test,P<0.05 is statistical meaningful.We use ROC curve to detect the sensitivity and speciality in diagnosing cancer development and relapse by Tim-1/2/3.We use Logistics analysis to detect the affecting factors of carcinoma development and relapse.Results Compared with benign ovarian tumor,expression of Tim-1/2/3 mRNA increased(P<0.05).The earlier the carcinoma relapses,the higher expression of Tim-1/2/3(P<0.05).The logistics analysis shows expression of Tim-1/2/3 is tightly correlated with relapse of ovarian carcinoma.The area below ROC curve is Y(0.909)>Tim-1(0.893)>Tim-2(0.888)>Tim-3(0.879).The value of Tim-1,Tim-2,Tim-3 and Y is 0.685,0.715,0.845 and 219.420 respectively,which shows combining testing of Tim-1/2/3 has good value in predicting development and relapse of epithelial ovarian carcinoma.Conclusion Tim-1/2/3 mRNA and protein expression may contribute to development and relapse of epithelial ovarian carcinoma and the combined detection of Tim-1/2/3 expression could be an effective method to predict the development and relapse of epithelial ovarian carcinoma.
