Study Based On Literature Review And Data Mining Approach To Analyze The Usage Patter Of Tibetan Medicine To Treat HAPC And Clinical Trial Of Duoxue Kang

Objective1.Through the ancient and modern literature research to investigate the etiology,treatment,diagnosis,and therapeutic principals of the disease,in addition,by using bibliometrics and knowledge map to analyze the research institutes,research field,research trends and distribution of researchers for high altitude HAPC.2.By using data mining approach to analyze the prescription compatibility law such as the high frequency medication mode,core drug combination,drug pair and new prescription of HAPC in Tibetan medicine,in order to define the inner relationship between“Disease-Prescription-Drug” and the medication regularity.Meantime,by using network pharmacology approach to explore the regulation mechanism of multi-component,multi-target,multi-channel of the four Tibetan medicine herbs of high frequency usage in the treatment of HAPC,and discovered the constructing the network model of "compound-target-pathway".3.Study the efficacy of Tibetan medicine Duo Xue Kang capsule in the treatment of HAPC by the clinical trial,providing scientific basis for the clinical application of this drug.Method1.Using the method of literature review unearths the pathogenesis,therapeutic principle and method,diagnosis and treatment of the HAPC from the ancient books or modern literature.Retrieves literature about HAPC from the database of the full text(CNKI)and Web of Science,taking advantage of the analysis function comes with the database,Note Express software and drawing and statistical functions of Excel to account annual changes,statistical research institutions and distribution of leading placebo researchers.Import Cite Space V software,adjust the time limit,the type of nodes and pruning items,etc.,to get the analysis Atlas of literature,keywords,periodicals.2.Collects drug information of HAPC treatment from literature database as ancient books of Tibetan medicine,combines with clinical status of treatment of HAPC in Tibetan medicine closely,and uses TCM inheritance auxiliary system(V2.5)software and software entropy integrated method,to explore the association rules of for the treatment of high altitude polycythemia(HAPC)and high altitude pulmonary edema(HAPE)in the Tibetan medicine,digging out administration mode of high frequency and the core drug combination,analyzing the medication regularity,defining the inner relation between “Disease-Prescription-Drug”.3.This paper digs out the four Tibetan medicine herbs of high frequency usage in the treatment of HAPC by the use of TCM inheritance auxiliary system(V2.5),screens the effective constituents of the four herbs through the literature and the use of TCMSP database,screens potential targets of each medicine by the use of Pharm Mapper server and Drug Bank database,obtains the corresponding path information with importing the obtained targets into the MAS3.0 database,carrys out the path notes and GO enrichment analysis with the combination of KEGG.constructing the network model of "compound-target-pathway" using the Cystoscope 3.4.0 software of the four herbs.4.Collects 100 cases Based on international Diagnostic Criteria of HAPC from Zêku County Tibetan Hospital in Qinghai Province Huangnan city,selects 14 male patients and 10 healthy volunteers as the research object in accordance with the relevant requirements of test,obtains the data of cerebral imaging of nuclear magnetic resonance,blood routine,signs and pulse oxygen saturation of blood of the treatment group before and after taking the Tibetan medicine "Duo Xue Kang Capsule" by the measure of intravenous perfusion scanning.Processes and analyzes the value of r CBV to the nuclear magnetic resonance imaging data through the data analysis method and Matlab5.3 software.Compare the difference of the indexes before and after taking the medicine,and finally to verify the clinical effect of Tibetan medicine "Duo Xue Kang Capsule".Result1.The author sorts out the ancient books and modern literature of Tibetan medicine,coming to the conclusion that this disease is caused by outside body digestion,absorption and on diet,drug and treatment method.Forms three major institutions of General Hospital of Tibet military area command,Qinghai People’s Hospital and Third Military Medical University,simultaneously a cooperation network of the of serum erythropoietin(EPO)with HAPC of Li Suzhi,Mou Xinbing,and Xue Zengjun,through the combination of literature metrology and knowledge mapping.Study on the foreign saussureae(87),red flower(82),Yuganzi(76),nutmeg(69)in the prescription for treating HAPC;Setting the number of support to 34,the confidence level 0.9,the use of the frequency of the first 5 drugs in turn are chebule-Yuganzi,clove-chebule,red flower-chebule,chebule-nutmeg,clove-nutmeg;The related drugs in common drug combinations are Maohezi→chebule,Maohezi→Yuganzi,Maohezi and Yuganzi→ chebule,chebule and Maohezi→Yuganzi,Guangzao→nutmeg.On the basis of core combination extraction,using unsupervised entropy hierarchical clustering algorithm,discovers that when the association rules Guangzao→nutmeg,nutmeg and Yuganzi→chebule,chebule and Maohezi→Yuganzi,concretio silicea bambusae and Bezoar→red flower,aloes and Guangzao→nutmeg.obtains21 new prescriptions by using unsupervised hierarchical clustering algorithm on the basis of the core combination extraction.3.Based on the collecting information of HAPC,the clinical status and practice of HAPC and Chinese medicine transmission support platform(V2.5)system software,five kinds material of Tibetan medicine Myrobalan、Woody、West safflower、Phyllanthus emblica(Which has been related to Phyllanthus emblica research,not repeated here)、nutmeg was found highly used during the treatment of HAPC prescription,which was obtained by the association rule and complex system entropy clustering method.Four kinds of chemical constituents were selected as the research objects from above material.TCMSP,KEGG,MAS3.0,Drug Bank,Pharm Mapper and Cytoscape software were used to study the effects,the target and mechanism of those four chemical constituents for the reatment of HAPC.It was found that the target 16、20、20、15 has relationship with HACP and the route17,17,24,14 has significant effect(for P<0.01).Through the analysis of the target and pathway,suggesting that the four herbs in the treatment of HAPC process,mainly through the promotion of cell cell proliferation and oxidative stress,play an anti-inflammatory effect,involved in regulating blood vessels and other functions,to reduce hemoglobin,Enhance the effectiveness of the body immunity to play a role in the treatment of HAPC.This provides a basis for further study of the mechanism of action of various herbs,and also provides scientific ideas and methods for the study of Tibetan medicine.4.The results of Blood routine Nuclear magnetic resonance brain imaging data and Hemodyna mics from three groups(Before taking medicine,after taking the medication,Normal control)are sig nificant.According to the ease of headache,dizziness,Purpura,asthma,palpitations,Hand and foo t fever of 14 person after they used Tibetan medicine called Du Xuekang capsule and the data oft heir Blood routine Sp O2 and PR,It is found to be effective in 13 cases,Ineffective in 1 case,the totaleffective rate was 92.9%.It also shows that the medicine do have effect on HAPC because the statistical result is significant(p<0.01).The conclusion also can be proved by the data CBFetc.,nucl ear magnetic resonance imaging(MRI)of the left frontal lobe,the right frontal lobe,the left temp oral lobe,the right temporal lobe,the left occipital lobe and the right occipital lobe of the patients.Conclusion:1.The literature review concluded that HAPC is a western disease and it is called "Chapei" in Tibetan medicine,it caused by increasing of bad blood,there are three types of syndromes such as wind,bile and phlegm types.2.The data mining found that there are similarities and differences of frequent herbs,prescription models,and core ingredients for HAPC and HAPE.3.The network pharmacology discovered the characteristics of the multi-component,multi-target,multi-channel of the Tibetan medicine for regulating systems.4.Clinical trial concluded that Tibetan formulae Dukukang capsule is effective for treating HAPC.Innovations:1.Firstly combined bibliometrics and knowledge map to analyze the trends of studying in HAPC2.Newly used data mining and network pharmacology to analyze the association rules and mechanism for Tibetan herbs.3.Firstly used the MRI techniques to explore the clinical efficacy of DUOXUEKANG capsule to treat HAPC.
