| Objective:Colorectal cancer is one of the most common malignant tumors of the digestive tract in the world.It has a high morbidity and mortality and has become a malignant disease that seriously endangers human health.The traditional Chinese medicine method has also played a key role in regulating and improving the internal environment of patients with its unique means of syndrome differentiation.This study was to investigate the effects of JXF on the biological behavior of colon cancer cells in hypoxic microenvironment.Methods:Human colon cancer SW620 cells were cultured in vitro and induced by hypoxic environment.The two groups were induced to culture under hypoxic conditions.PCR and Western Blot were used to detect the expression of HIF-1ɑ,VEGF,MMP-9,survivin,Bcl-2 and Caspase-3 m RNA and protein in colon cancer cells,cells were treated with 6 different 5-Fu concentrations.SW620 cells in logarithmic growth phase were enlarged to 2 culture dishes.They were randomly divided into JXF and control group.Both groups were induced to culture in hypoxic environment.PCR and Western Blot were used to detect the expression of HIF-1ɑ、VEGF 、 Bcl-2 、 Caspase-3 、 MMP-9 、 survivin;MMT was used to detect cell proliferation;Transwell was used to detect cell migration and invasion ability;Flow cytometry was used to detect cell apoptosis,and different 5-Fu concentration groups was used to detect cell survival rate.Results:(1)Compared with the control group,the levels of HIF-1ɑ,VEGF,MMP-9,survivin and Bcl-2 m RNA and protein in the SW620 cells were increased,and Caspase-3 m RNA and protein were decreased;the cell viability of SW620 cells in hypoxic group increased at 48,72 and 96 h,the number of penetrating ventricular cells increased,and the apoptosis rate decreased.When the concentration of 5-Fu was 4,8,16 μmol/L,the survival rate of SW620 cells in hypoxia group increased.(2)Compared with the control group,the m RNA and protein levels of HIF-1ɑ,VEGF and Bcl-2 in SW620 cells of JXF group decreased,Caspase-3 increased;the survival rate of SW620 cells decreased at 48,72 and 96 h,the number of migrating and penetrating ependymal cells decreased,and the apoptotic rate increased in the JXF group,while the survival rate of SW620 cells in the JXF group decreased when the concentration of 5-Fu was 4,8 and 16μmol/L.(3)The tumor volume and weight were changed in JXF groups as compared with the model group.Conclusions:JXF Decoction can down-regulate the expression of HIF-1ɑ in colon cancer cells in hypoxic microenvironment,indirectly affect the expression of its downstream target genes,inhibit the proliferation of colon cancer cells,promote cell apoptosis,reduce cell migration,and enhance the sensitivity of colon cancer cells to chemotherapeutic drugs,so as to play an anti-cancer role. |
