Allogeneic full-thickness cartilage transplantation is gradually considered to be the preferred clinical solution for the treatment of full-thickness cartilage damage during recent years.It is to provide biological active bone and cartilage tissues,as well as applying extensive,even specific shaped,customized repairment to the lesioned area.However,the immunological rejection caused by T cell mediation,involving multiple inflammatory cells and factors,leads to Numerous surgical transplants fail.Thus,it is critical to avoid immunological rejection in order to ensure the success of Allogeneic full-thickness cartilage transplantation.In regards of the challenge of immunological rejection,the conventional methods,such as decellularizing and deantigenic treatment to the grafted bone and cartilage tissues,not only potentially eliminating the biological activity,but also incapable of avoiding immunological rejection radically.It is difficult to achieve the full-thickness cartilage repairment.This experiment implanted the agarose,which has the basic fibroblast growth factor(bFGF),into the recipient rats to form an immune encapsulation locally.Hence to escape the host immune mechanism.This experiment also discusses the mechanism of immunological rejection,disease course evolution and according avoiding methods.Accordingly,it would provide a new method of full-thickness cartilage repairment.Followed are the main content and achievements:1.In vitro cell experiment,it is found that appropriate concentration of bFGF could promote the proliferation and differentiation of rat chondrocytes in vitro.The optimal concentration range of bFGF is 10 ng/mL-100 ng/mL,once the concentration of bFGF reached 500 ng/mL,the promotion effect begins to decrease and the inhibition appears,which provides a theoretical basis for the application concentration of bFGF solution in vivo.2.In this study,through in vivo experiments about the strength of acute immune response at different stages after full-thickness cartilage allogeneic/allogeneic transplantation.The intensity of the reaction peaked 5 days post-surgery,and then gradually fell back.Meanwhile,the body completed the attack on the allogeneic implant through this process.3.Seven days before the operation of full-thickness cartilage defect allotransplantation in rats,preimplantation of bFGF/agar-agar gel column complex was performed,which could effectively reduce the inflammatory response of rats,protect the second-stage graft,and promote whole-layer cartilage allograft repair.It provides a new method and thought for the whole layer cartilage repair.In conclusion,this study designs and constructs a series of surgical,as well as interfering methods,to protect the grafted bond and cartilage tissues,which promote the full-thickness cartilage repairment.
