Objective:To explore the role of miRNA-regulated inflammatory cytokine IL-17 A in postmenopausal osteoporosis.Analyze the targeted regulation effect of hyperoside,a traditional Chinese medicine component on postmenopausal osteoporosis.Elaborating the pathogenesis of postmenopausal osteoporosis from the perspective of epigenetic immune regulation.Provide new targets and experimental basis for clinical treatment.Methods:1.Use bioinformatics software and chip to predict the upstream miRNA of IL-17A;The dual luciferase reporter gene system verifies the binding site.2.Induce osteoclasts in vitro and observe the effect of transfection-regulated miRNA expression on the function of osteoclasts after IL-17 A expression.3.Establish a postmenopausal osteoporosis model to observe the effect of regulating miRNA on IL-17 A expression and osteoporosis.4.Observe the effect of hyperoside on miRNA and IL-17 A and its regulating effect on osteoporosis.Results:1.miRNA-19a-5p can bind to the 3'UTR of IL-17 A,thereby regulating IL-17 AmRNA and protein expression.2.Decreased expression of miRNA-19a-5p can up-regulate the expression of IL-17 A,and then increase the osteoclast marker protein.3.Regulating miRNA-19a-5p can regulate the expression of IL-17 A mRNA and protein in mice,and then affect the changes of bone mass in the postmenopausal osteoporosis model.4.Hyperin can inhibit the expression of IL-17 A by increasing miRNA-19a-5p,and increase the bone mass of postmenopausal osteoporosis models,and play a role in bone protection.Conclusion:1.IL-17A-mediated inflammation is involved in the pathological process of postmenopausal osteoporosis.2.miRNA-19a-5p regulates the expression of IL-17 AmRNA and protein by binding to IL-17 AmRNA3'UTR,thereby regulating the inflammatory response and cause bone loss.3.Decreased expression of miR-19a-5p can lead to increased IL-17 A,which may be an important cause of osteoporosis.4.Epigenetic immune regulation is involved in the occurrence of postmenopausal osteoporosis.Targeted regulation of miR-19a-5p/IL-17 A axis may become an effective way to prevent and treat postmenopausal osteoporosis.5.Targeted regulation of miRNA-19a-5p/IL-17 A may be one of the key mechanisms of hyperoside in the treatment of postmenopausal osteoporosis;this mechanism enriches the theory of "deficiency of kidney essence" in TCM,and provides verification for TCM theory from a microscopic perspective.
