| Objective:To observe the effects of Taohe Chengqi Decoction on the renal function,blood routine,renal pathological changes and Wnt/β-catenin signaling pathway in rats with chronic renal failure(CRF).The mechanism of action provides ideas and objective experimental evidence for the clinical treatment of CRF.Method:①Modeling and grouping:100 male Wistar rats of clean grade,7 weeks old,weighing 180-200g,purchased for 1 week after adaptation,randomly divided into blank group,sham group,There were 25 rats in the model group and Taohe Chengqi Decoction treatment group(treatment group).An experimental animal model was established using a 5/6 nephrectomy procedure.For the first time,the left kidney 2/3 resection was performed in both the model group and the treatment group.The second operation was performed 7 days later and the right kidney was removed.Both penicillin injections were given intraperitoneally for 3 consecutive days.The blank group was not treated.The sham group only had a double kidney capsule peel.The blood venous plexus of each group was collected for blood creatinine three weeks after operation and the success of the model was observed.②Treatment and administration:4 weeks after surgery every morning:the treatment group to Taohe Chengqi Decoction gavage;blank group,sham operation group and model group were given equal volume of saline gavage.The doses used were all converted to 6 times the dose per kilogram of body weight in clinical adults,and the administration time was 8 weeks.From the beginning of the administration,the general condition of the rat was observed and recorded every day,and the body weight was measured once a week.The rats died during the experiment were recorded and the cause of death was analyzed.③Observation index:8 weeks after the administration of the drug,blood was collected from the abdominal aorta of the rat under anesthesia for detection of blood routine and renal function of the rat;the rat kidney was probed after the blood was collected,and the residual kidney tissue was cut to In two parts,a part of renal tissue was placed in a cryotube,Western blot technique was used to detect the protein contents of Wnt3a,Wnt5a,Wnt5b,Wnt8b,Wnt9b,β-catenin,and E-cadherin in renal tissue.The other part of the kidney tissue was stored in poly In the formaldehyde solution,the pathological changes of each group of kidneys were observed with a light microscope after treatment.Result:①Compared with blank group and sham operation group,the contents of RBC and Hb in the model group and the treatment group were significantly lower,the contents of serum Scr and BUN were significantly increased(P<0.05),and the Wnt3a,Wnt5a,and(3-catenin protein contents in the kidney tissue were significantly increased.Significant increase(P<0.05),Wnt5b,E-cadherin protein content was significantly reduced(P<0.05),Wnt8b,Wnt9b protein content was no significant difference(P>0.05),light microscopy glomerulosclerosis,interrenal Significant fibrosis;②Treatment group compared with the model group,RBC,Hb content increased significantly,Scr,BUN content decreased significantly(P<0.05),renal tissue Wnt3a,Wnt5a,β-catenin protein content decreased significantly(P<0.05),Wnt5b,E-cadherin protein content was significantly increased(P<0.05),Wnt8b,Wnt9b protein content was no significant difference(P>0.05),renal fibrosis under light microscope than the model group.Conclusion:① Taohe Chengqi Decoction can significantly improve renal function in CRF rats,correct anemia,and delay the progression of renal fibrosis;②The mechanism of Taohe Chengqi Decoction in treating CRF may be modulating the Wnt family of factors to inhibit the expression of Wnt3a,Wnt5a,β-catenin proteins and promote the expression of Wnt5b and E-cadherin proteins. |
