Medicine Of Taohechengqi Decoction On Wnt/beta-catenin Signaling Pathway Intervention On Renal Interstitial Fibrosis In Chronic Renal Failure Rat

Objective:Through the animal experiment to observe Taohechengqi Decoction on chronic renal failure(CRF)rats with renal fibrosis Wnt/beta-catenin signal pathway intervention effect,feasibility analysis of Taohechengqi decoction delay CRF,investigate its mechanism,so as to provide clinical ideas and experimental basis for the treatment of CRF.Methods:5/6 nephrectomy animal model CRF rats,60 male 7 week old Wistar,weight 180-220g rats of clean grade,repurchase after adaptive breeding for 7 days,were randomly divided into control group,sham operation group,model group,Niaoduqing group,Taohechengqi decoction treatment group with 12 rats in each group.White group without surgery,the sham operation group underwent bilateral renal capsule stripping,model group,control group and treatment group were treated by 5/6 nephrectomy and intraperitoneal injection of penicillin for 3 days.After 28 days the tailvein after detection of serum creatinine(SCr),blood urea nitrogen(BUN)detection if the model is successful,then every morning at about 9 different drugs administered daily for 56 days,The general situation of rats in each group was observed and recorded,and the number and causes of death were analyzed.Fifty-seventh days in sterile conditions under the abdominal aorta Arterial blood,blood serum in the detection of red blood cell(RBC)count,hemoglobin(Hb)content,the renal function of SCr,the content of BUN,using immunohistochemical method to detect Wnt4 in renal tissue of Wnt5a,Wnt5b,protein content of beta-catenin,rats were sacrificed and renal tissue,Hematoxylin-Eosin Staining for pathological observation of light mirror.Results:1 compared with the blank group,the count of RBC rats in the model group,the content of Hb was decreased,SCr,BUN were significantly increased(P<0.05),Wnt4,Wnt5a,the content of beta-catenin was significantly increased,Wnt5b content decreased significantly(P<0.05),model group of rat renal tissue morphology mostly disappear mirror,cell shrinkage or uneven distribution,glomerular sclerosis,renal tubular dilatation or atrophy,renal interstitial fibrosis,inflammatory cell infiltration;2 compared with rats in model group,control group and treatment group rats RBC count,increasing the content of Hb,SCr,BUN content decreased(P<0.05),Wnt4,Wnt5a,reduce the content of beta-catenin,Wnt5b content increased significantly(P<0.05),light degree of hyperplasia of fiber small sclerosis and renal kidney between the two groups was significantly lighter than the model group,and taohechengqi decoction is more effective than Niaoduqing.Conclusion:1 The general situation of taohechengqi decoction can improve CRF rats to a certain extent,to correct the anemia status,with elevated RBC count and Hb content,but also can improve the renal function,to reduce the serum SCr,BUN content;2 taohechengqi decoction can affect Wnt/beta-catenin signaling pathway inhibition of Wnt4,Wnt5a,beta-catenin protein and promote the expression of Wnt5b protein,combined with renal pathological changes under light microscope,we can get the effective renal fibrosis damage,thus delaying the progression of CRF.
