| Objective:Traditional Chinese medicine plays an important role in the prevention and control of diseases,forming a unique theoretical system.Its theory and principles of epidemic treatment urgently need to be further sorted,excavated,and improved.This project uses methods such as literature research,data mining,and experimental verification,taking ancient medical works as the research object,combined with the “uncloging and exuding”dispelling evil method to explore and organize the theory and rules of traditional Chinese medicine for epidemic treatment,inheriting the academic ideas of ancient medical practitioners,summarizing the basic laws of traditional Chinese medicine for epidemic treatment,screening effective Chinese medicine for epidemic treatment,and then scientifically verifying the effectiveness and mechanism of effective prescriptions for epidemic treatment through experimental research.This study scientifically summarizes the experience and principles of traditional Chinese medicine in treating epidemics,which can provide scientific basis for optimizing traditional Chinese medicine treatment plans for influenza and other diseases today.Methods:1.Literature research: Combining manual retrieval with electronic retrieval,sort out literature materials related to the "transparent" dispelling evil method,and search,organize,and analyze them in chronological order.2.Data mining: Using the literature research section as the data source,search for the“uncloging and exuding” dispelling evil method’s formula that meet the inclusion criteria,use Microsoft Office Excel 2021 to input the required information for the study,standardize the information,and establish a database.Using the ancient and modern medical case cloud platform for frequency statistics,cluster analysis,and complex network analysis,we can obtain the syndrome and treatment laws of medical practitioners in the past,and explore the core prescription of the "transparency and elimination of evil" method for treating epidemics.3.Animal experiment: Establish a mouse infection model of type A influenza virus,and evaluate the “uncloging and exuding” dispelling evil method’s formula based on this.Evaluate the protective effect of influenza virus infected mice by monitoring their weight changes and mortality within 14 days.In addition,infected mice were euthanized at random on the 3rd day(3 dpi)and 5th day(5 dpi)after infection,and their lung tissues were collected for lung index measurement and HE staining analysis.The m RNA levels of representative cytokines and chemokines in lung tissue homogenate were detected using RT q PCR,and the protein levels in their anti influenza virus related pathways were detected using Western Blot(WB).Results:1 The epidemic treatment concept of the “uncloging and exuding” dispelling evil methodThe method of “uncloging and exuding” dispelling evil has long been reflected in the treatment of diseases,first seen in the Neijing.It is based on the location of the disease,guided by the principle of expelling evil nearby,and aimed at eliminating evil.It includes various treatment methods such as sweating,descending,penetrating,and vomiting.The results showed that the application of the “uncloging and exuding” dispelling evil method in the "cold epidemic" is based on the transformation of non seasonal cold from the surface to the inside of the "three yang(surface)-three yin(interior)",based on the location of evil energy retention,using the six meridians and six parts as transmission channels,and applying treatment methods such as sweat,vomiting,descending,and penetration to“uncloging and exuding” dispelling evil.The application of the “uncloging and exuding”dispelling evil method in "Wendu" is based on the principle of non seasonal warmth,dormancy and subsequent onset,and external penetration of the skin surface.Therefore,it is treated by external penetration and internal purging of heat and toxins.The application of the “uncloging and exuding” dispelling evil method in epidemic prevention is based on the direct accumulation of epidemic qi from the mouth and nose on the membrane,middle jiao,and blood,with the transformation mode of going out of the surface,going in,and going out of the surface and going in.It provides two exothermic pathways for the “uncloging thirst;Common high-frequency drugs include Scutellaria baicalensis,rhubarb,and gypsum,in addition to the auxiliary medicine licorice;The top three flavors of the medicine are bitterness,spiciness,and sweetness,while the top three meridians of the medicine are the lungs,stomach,and spleen.The clustering analysis results show that the overall database of diseases is divided into 9 groups,namely gardenia and fermented soybean;Huanglian and Zhimu;Rhubarb and mirabilite;Mahuang and Guizhi;Poria cocos;Rhinoceros horn,cohosh hemp,Ophiopogon japonicus;Ginger and ginseng;Gypsum,kudzu root;Red peony,Chaihu,Licorice,and Scutellaria baicalensis;The cold epidemic data is divided into 8groups,which are almonds;Mahuang and Guizhi;Soy beans,gypsum,kudzu root;Scutellaria baicalensis,gardenia,rhubarb,and mirabilite;Ginseng,ginger;Poria cocos and Fructus Aurantii;Chaihu,Chishao;Rhinoceros horn,cohosh,licorice,and Ophiopogon japonicus;The epidemic database is divided into 10 groups,namely gypsum and ephedra;Gardenia and Phellodendron;Houpu and Zhishi;Rhubarb and mirabilite;White peony and Chaihu;Qianghuo;Licorice and Zhimu;Angelica sinensis,native place;Platycodon grandiflorus;White stiff silkworm,cicada molt,Scutellaria baicalensis,Coptis chinensis.Through complex network analysis,three core drugs were obtained: one is Scutellaria baicalensis,gypsum,Ophiopogon japonicus,Rhubarb,Cimicifuga,Gardenia jasminoides,Glauber’s salt,Chaihu,Kudzu root,and Licorice;The second is Scutellaria baicalensis,Ophiopogon japonicus,Gypsum,Cimicifuga,Pueraria lobata,Gardenia jasminoides,Rhubarb,Chaihu,Chishao,and Licorice;The third is Scutellaria baicalensis,Coptis chinensis,White Silkworm,Cicada molting,Rhubarb,Glauber’s salt,Gypsum,Magnolia officinalis,Zhimu,and Licorice.3 Research on the protective and anti-inflammatory mechanisms of the “uncloging and exuding” dispelling evil method’s formula on influenza virus infected miceBy analyzing the weight changes and mortality of mice within 14 days of infection,it was found that the core prescription(Hexin Chufang)of the “uncloging and exuding”dispelling evil method can protect mice from weight loss and reduce mortality.Through pathological examination of mouse tissues,it was found that after infection with influenza virus,the lung index of mice increased,pulmonary edema occurred,and pathological damage such as blood stasis appeared.A large number of lymphocytes and macrophages were visible in the alveolar cavity,and the structure of the alveoli was unclear in many places,with many necrotic cell fragments appearing.Large areas of alveolar walls showed infiltration of granulocytes.The effective prescription of the “uncloging and exuding”dispelling evil method can protect the pathological damage caused by influenza virus to lung tissue.Moreover,compared with influenza virus infected mice,the effective formula of "clearing through and dispelling evil" significantly reduced the m RNA expression of cytokines IL-6,TNF-α and IL-1 β,interferon IFN-α and IFN-γ,as well as chemokines CCL-3 and CXCL-10 in the lung tissue of influenza virus infected mice.For the potential inflammatory response mechanism,it was found that MyD88 was activated in virus infected mice and had an impact on downstream NF-κ B signaling.The effective formula of "transparency and elimination of evil" showed inhibition of MyD88 and p65 protein expression,and activation of I κ B protein expression.Conclusion:This study scientifically summarizes and proposes the concept of “uncloging and exuding” dispelling evil method and promoting the rule of law in epidemic prevention.The“uncloging and exuding” dispelling evil method is based on the physiological principle of "the human body is not limited to the exterior and interior",integrating the pathogenesis and transmission laws of different types of evil qi.According to the different disease locations,corresponding treatment methods are used to eliminate the evil nearby.This study summarized the medication rules of the “uncloging and exuding” dispelling evil method for treating the epidemic and screened out the core prescriptions for treating the epidemic.Through data mining,it was found that the pathogenesis of the epidemic is mainly due to pathogenic heat stagnation,and the medicinal taste is mainly composed of hard work and sweet taste.The medicinal meridians are mainly focused on the lungs,stomach,and spleen meridians.In terms of application,they mainly focus on relieving muscle,penetrating the surface,and promoting diarrhea and excess evil.Gypsum,Scutellaria baicalensis,and rhubarb were selected as the core prescriptions(Hexin Chufang).The core prescriptions(Hexin Chufang)screened in this study has a protective effect on influenza virus infected mice,and can positively regulate the m RNA expression of cytokines,interferon,and chemokines in the lung tissue of influenza virus infected mice.Its potential mechanism may be related to the regulation of the MyD88/NF-κ B signaling pathway. |
