A kind of endophytic fungi has been isolated from 12 strains of endophytic fungus of Huperzia serrta (Thunb.) Trev. with the target as the yield of huperzia A. The endophytic fungus was then identified by molecular methods. The biological characteristics and fermentation condition have been studied. The results were as follow:1. No. 190 strain was isolated from 12 strains endophytic fungus of Huperzia serrta (Thunb.) Trev. as it might have the component huperzine A or the analogue of huperzine A analyzed by TLC, HPLC and HPCE(high perform capillary electrophoresis).2. Wet-room culture method was used to observe the microstructure of No. 190 strain and the pictures of chlamydospores, spores and mycelium were taken.3. No. 190 strain was identified by ITS sequence analysis and the result showed that it belongs to Podospora sp.. By single factor analysis, the optimum culture medium included: glucose as the carbon source, yeast extraction as the nitrogen source and natural pH.4. No.190 strain fermentation biomass model was established by ANN(artificial neural network). In this study, UAER (Uniform Analysis of Experimental Result) was first proposed and applied so that the workload was reduced greatly and the results were reliable.5. Huperzine A content in No. 190 strain fermentation products was tested by HPLC and the results indicated that there were Huperzine A or the analogue of huperzine A but the content was too small to obtain the huperzine A monomer at the moment.6. The components of the phase of petroleum ether of the mycelium of No. 190 strain and of the fermentation fluid were analyzed by GC-MS. 60 compounds in total were detected (30 peak groups for the mycelium and another 30 for the fermentation fluid). The results showed that there were great differences in contents and types of compounds between the mycelium and the fermentation fluid.7. Antitumor activities of petroleum ether extraction and chloroform extraction of the fermentation fluid and the mycelium of No. 190 strain were detected. The conclusions are that the antitumor activity of the fermentation fluid was much better than the mycelium by using petroleum ether as extracting solvent but the antitumor activity of the fermentation fluid was much better than the mycelium while by using chloroform as extracting solvent. There will be more study for mechanism.
