Succus bambosae is the yellow and bright juice excreted from the baked fresh stem of Phyllostachys nigra (Lodd.) Munro var. henonis (Mitf) Stapf ex Rendle and other kinds of bamboo. Succus bambosae has the functions of removing heat-phlegm and arresting convulsion. It can be applicable for treatment of cough and asthma due to phlegm-heat, infantile convulsion due to phlegm-heat, apoplexy and abundant expectoration, and other syndromes.In order to solve some actual problems, such as: unification in preparation process, lower extraction rate, large waste of original plant, serious pollution of environment. The Phyllostachys nigra (Lodd.) Munro var. henonis (Mitf) Stapf ex Rendle was choiced as raw material in this study. The comparative researches was made on different preparation process to produce Danzhuli and its quality by UASLG (Ultraviolet Absorption Spectra Lines Group) and HPLC (High Performance LiquidChromatography).1. Established the factors and levels in the orthogonal designs for percolation by researches on single factor. Using content of gross amino acid and gross hydroxybenzene as evaluating indexes, we selected the optimal percolating conditions by the orthogonal text, which were as follows:18 hours' soaking, percolating velocity (Mml'rnin"1 with tenfold 60% ethanol. The contents of gross amino acid and gross hydroxybenzene processed by this method are 176.897mg/100g and 20.102mg/100g.2. Established the factors and levels in the orthogonal designs for refluxing method by researches on single factor. Using content of gross amino acid and gross hydroxybenzene as evaluating indexes, we selected the optimal extraction conditions by the orthogonal text, which were as follows:60 minutes' soaking, tenfold 70% ethanol, refluxing two times, each time for two hours. The contents of gross amino acid and gross hydroxybenzene processed by this method are 330.686mg/100g and 34.012mg/100g.3. The optimal preparation process was selected by comparative researches on different four backed methods, which were refluxing method. This method changed the unenlightened conditions of traditional baked process, greatly economizing the raw materials, ensuring theproductivity of Succus bambosae. It can be used in industrialized production.4.Using UASLG to identify Danzhuli obtained from different four preparation methods and Xianzhuli oral liquids bought from pharmacy. This method is external and exact. It can reflect macroscopical traits of components in samples and can identify Chinese traditional medicine effectively.5. The method for determining the content of guaiacol in Danzhuli produced by refluxing method was established. It provides foundation for farther researches on quality standard.
