In this dissertation, preparative chromatography method of purifying saponins inpanax notoginseng leaves was developed. The optimal technical parameter ofpreparation procedure was determined. The fingerprints of saponins in panaxnotoginseng leaves and the chromatogram preparation were established to controlcriterion for the modernization of traditional Chinese medicine.Reverse-phase liquid chromatography had been hot issues in separation of the natureproducts. Using the crude extracts of stems and leaves as material in preparativeliquid chromatography column, the effects of the flow rate, the contents of the mobilephase and the sample quality in the separation process were investigated respectively.Thus, the suitable chromatography conditions were determined. The content ofginsenoside Rb3 in the product was about 80%~90% and the recovery yield was 70%.This method was effective to purify saponins in panax notoginseng leaves.The fingerprints were one of the available techniques to the quality control of it.HPLC and LC/MS were used to establish the fingerprints of saponins in panaxnotoginseng leaves and the chromatogram preparation. The analysis of mass spectrumshowed that: the chromatogram preparation had ginsenoside Rb3. The experimentmade the high-quality, controllable, stable and uniform fingerprints to overall reflectthe quality of saponins in panax notoginseng leaves and the chromatogrampreparation.
