The Research On The Extracting Of Diosgenin From Dioscorea Zingeberensis Wright

Dioscorea zingeberensis wright is a special plant of China, used to produce steroid drugs. An effective content is diosgenin from it. In order to increase the extracting rate of diosgenin, we studied all the factors that affect diosgenin yield in the process of fermentation, hydrolysis and extraction.In sucession, we studied the purpose ofα-amylase, fibrin enzyme,xylan enzyme andγ-amylase on the increasing of extracting rate of diosgenin.And we first made use of RP-HPLC to indentify the quality of diosgenin.The result is that the diameter of the tuber of Dioscorea zingiberens affect remarkably the extracting rate of diosgenin.When pH of substance is close to 7,the extracting rate of diosgenin is maximal.On the basis of single and orthogonal test,the optimum technological condition was material diameter≤1mm, fermentation temperature 20℃～40℃, fermentation time 24h, acid concentration 2.5mol/l, acid hydrolysis time 6h., circumfluence time 12min, extracting time 6h.The result of ways of enzyme is thatα-amylase increased the extracting rate of diosgenin by 11.59%, and reduced draff yield by 2.88%, and had no impact on the quality of diosgenin. Fibrin enzyme increased diosgenin yield by 5.46%, and reduced draff yield by4.72%, and did not affect the quality of diosgenin. Xylan enzyme increased the extracting rate of diosgenin by 1.02%, and reduced draff yield by 6.02%, but it had bad impact on the quality of diosgenin. The result of ways of enzyme is thatγ-amylase had no impact on diosgein yield and draff yield.The ways of bathing did not work on diosgenin yield.Through synthetical comparability, we can increase yield of diosgenin by crushing dmaterial diameter≤1mm, flushing substance close to 7,appendingα-amylase to fermnt.And we can counteract it with Ca(OH)2before bathing to save abundant water.
