| In this paper, the resveratrol extraction from grape skin by technology of organic solvent surge extraction, supersonic extraction technology and microwave-assisted extraction technology were studied, and utilize macroporous adsorption resin to purify resveratrol, and the various techniques parameters of the three extraction methods were selected and optimized. HPLC analysis sample resveratrol were adopted in order to provide theory data for development of grape skin.The ethonal was used as extractor to extract resveratrol by organic solvent surge. Based on single fraction experiment, the orthogonal experiments were studied for the noticeable factors such as grinding granularity, extraction temperature, proportion of water to material, ethonal concentration and extraction time. The order as follows ethonal concentration > extraction temperature > the proportion of water to material > extraction time. From the arrange analysis and variance analysis, ethonal concentration effected the extract result of resveratrol and arrive at the most noticeable level. The results indicated that the extraction temperature 70℃, the proportion of water o .material 1:35, ethonal concentration 60%,solution pH4, extraction time 4h .On this condition,the obtaining rate is 0.165%.Resveratrol was processed and ethonal used as extractor by supersonic extraction technology for enhancing the extraction rate.Effect of several factors on supersonic extraction was explored using single fraction experiments and orthogonal experiments. The optimum technology conditions was selected, and the the results showed that the ethonal concentration60%, the proportion of water to material1:25, the time of work and intermission 1:2,the ultrasound power 500W. On this condition, the obtaining rate is 0.277%.From the arrange analysis and variance analysis, indicated that the order of five factors :the first is supersonic time the time of work and intermission. The second of ssupersonic power. The third of the proportion of water to material. The forth of ethonal concentration, and supersonic time was the most noticeable level..The Resveratrol was extracted and ethonal used as extractor by microwave-assisted extraction has been studied. Based on single experiment, the orthogonal experiments were studied for the noticeable factors such as microwave time, the proportion of water to material and microwave time. The order of three factors were microwave time > microwave power > the proportion of water to material. The results indicated that microwave time 70s, the proportion of water to material 1:20, microwave power 600W. On this condition, the obtaining rate is 0.284%.The research of three kinds of technology showed that: considering about the component and content of extract, solvent surge extract affected extract purification, and efficiency was low and wasted time. The other two kinds of technology that components were the same basically and efficiency was high. Three kinds of technology belong to chemical separate method of nonsolvent rudimental, and operation stable.Resin was used to purify Resveratrol,AB-8 model resin was optimum while the extract impact was considered. The sample speed of 1.5BV/h,concentration of 10mg/ml, pH 4, desorption concentration 80% ethonal and desorption speed of 0.75BV/h were the optimal conditions.After the Resveratrol was purified and detected by HPLC from the sample and criterion chart we know that peak model was the same basically. And the other peak maybe the interfere of a litter impurity. |
