Canarium album belongs to the Burevaceae and category of evergreen tree.The fruits are oval and green,which are also known as olive.It is a kind of plant resources which could be used for food and medicine.The effective active ingredient of the Canarium album is polyphenols which is antibacterial,antiviral,antipyretic,hepatoprotective and choleretic,immunomodulatory, anti-tumor,reduceing blood pressure,reducing blood fat,purging free radical and so on.In this subject,Canarium album produced in Chongqing was used as experimental materials,from which polyphenols were extracted and purified to measure the oxidation resistance.In this experiment,gallic acid were used as the standard material to identify the appropriate conditions of Canarium album polyphenols through Folin-Ciocalteu;with the microwave-assisted extraction method,the best method of Canarium album polyphenols extraction was determined through quadratic regression orthogonal rotary experiments;the best conditions for purification of Canarium album polyphenols with AB-8 macroporous resin was explored;compared with the common antioxidant,the antioxidant properties of Canarium album polyphenols purified with AB-8 resin were studied within the six systems:antioxidation of oil,ablity of reverting,removing of H2O2,free radicals·OH,superoxide anion(O2-·),nitrite(NO2-);some active ingredients of Canarium album polyphenols were determined through ultra-high performance liquid chromatography method.The results of the experiment are as follows:(1) Total centent of polyphenols was quantitatively determined by means of Folin-Ciocalteu reagent which formed a highly stable blue colour and with sufficient precision and ruggedness.In this paper,gallic acid was used as the standard substance,and there was a very good linearity when the concentration of gallic acid ranging from 0.05mg/mL~0.30mg/mL.The standard curve equation is Y=0.3309X-0.0237,and the R2 is 0.999.The RSDs of precision and recovery of the experiments are 0.938%and 0.404%.(2) Quadratic regression orthogonal rotary experiments for microwave-assisted extraction of polyphenols were used in this paper.And the results were analyzed by DPS.The results indicated that the 3 factors in the experiments,microwave-assisted extraction time(X1),microwave power(X2),solid vs.liquid ratio(X4),had strong influences on the extraction effects.And the interaction between microwave-assisted extraction time(X1) and solid vs.liquid ratio(X4) is also strong.The influencing extents of the 4 factors are:solid vs.liquid ratio(X4)>smicrowave-assisted extraction time(X)1)>microwave power(X2)>ethanol concentration(X3). The best extraction condition are:microwave-assisted extraction time is 50 seconds,microwave power is 600W,ethanol concentration is 70%,and solid vs.liquid ratio is 1:25.Using these conditions,we got 101.85mg polyphenols from 1g dry powers of Canarium album,and it is about 0.62%lower than the theoretical calculation value.The equation which showed the relationship between the extraction effect and the 4 factors is: Y=96.2571+4.05375X1+2.19958X2+4.67125X4-2.65731X12-1.08231X22-2.90731X32-2.59481X42 +1.64562X1X4(3) By captive tests,we detected the absorbing and eluting capacities of 5 kinds of macroporous absorbent resins produced by two companies.The results indicated that AB-8 resin was the best one with higher absorbing and eluting capacities than the other 4 kinds of resin.AB-8 resin could absorb polyphenols from the extraction quickly within 4 hours.The 70%of ethanol had a strong eluting effect to the polyphenols absorbed on the AB-8 resin,the eluting rate of polyphenols would arrive at 90.72%within 4 hours.Based on the absorption theory of AB-8 resin,this research studied on the contentration of ethanol and the speed of moving and washing.In the dynamic adsorbting and eluting experiments,the results analysis showed that:when the speed was 1.0mL/min and the sample concentration was 2.34~3.51mg/mL,the polyphenols adsorbting rate was the highest;when 70%ethanol used as eluent at the speed of 1.0mL/min,the elution peak was concentration and symmetry without obvious tailing phenomenon,and the polyphenols absorbed on 20g AB-8 resin could be eluted by 390mL 70%ethenol.(4)The purified Canarium album polyphenols were measured by UPLC,and the result presented that the content of Rutin which was 0.649mg/L.In the purified Canarium album polyphenols, flavonoids shared a larger proportion.(5) The antioxidant properties of Canarium album polyphenols extractions were studied through six different systems:edible lard,ablity of reverting,removing of H2O2,free radicals·OH, superoxide anion(O2-·),nitrite(NO2-).The results showed that,in the oil system,the anti-oxidation effect of 0.70g/kg of the Canarium album polyphenols extractions adding to lard was similar to that of 0.2g/kg of BHT.After 25 days' storage under the condition of 60℃,the peroxide value of the lard added 0.70g/kg of the Canarium album polyphenols extractions was 13.46meq/kg,which is lower than the national standard of 16meq/kg;the Canarium album polyphenols possesses strong reverting capacity,when the measured density ranged from 0.24 to 0.42mg/mL,the reverting capacity of Canarium album polyphenols would increase as the increase of the measured density.When the density was 1.96mg/mL,the A700 values of the reaction products of polyphenols was 1.84 times higher than that of BHT and 1.58 times than that of ascorbic acid;in the H2O2 system,the scavenging capacity of the Canarium album polyphenols extractions was almost similar to that of ascorbic acid,which was stronger than that of BHT. When the density of ascorbic acid was 0.98mg/mL,its purge rate reached 98.4 percent and extractions' maximum purge rate was 95.7%,while maximum purge rate BHT was only 84.5%;in the free radicals·OH system,the purge·OH rate of Canarium album polyphenols extractions in the reaction system was higher than that of ascorbic acid and BHT.When the density was 0.98mg/mL,the purge·OH rate of Canarium album polyphenols extractions was 1.20 times higher than that of ascorbic acid and 3.62 times than that of BHT;in the superoxide anion O2-·system,the scavenging capacity of Canarium album polyphenols extractions was weaker than that of ascorbic acid and BHT;in the nitrite system,the scavenging capacity of ascorbic acid is stronger than that of olive polyphenols.
