Poly (amino acid) has become a new kind of biodegrable polymeric materials due to good compatibility, especially poly (γ-benzyl-L-glutamate)(PBLG).The study on the new modified PBLG biomedical composite has allimportant significance because its disadvantages such as poor hydrophility,low degrability and non-functional group limit its biomedical application.The monomer of PBLG, N-carboxy-γ-benzyl-L-glutamate anhyride (Bz-L-Glu-NCA) was prepared throughγ-benzyl-L- glutamate (Bz-L-Glu) at first.In order to simplify the modification route and improve the yield ratio, the novel allyl-terminated poly (γ-benzyl-L-glutamate)(A-PBLG) was designed and successfully prepared using allylamine as initiator.A-PBLG have many advantages such as good biocompatibility and innocuity of degradation products and having the functional group(allyl) that improves its biomedical application.In order to improve the low biodegradation rate and the poor hydrophility of A-PBLG. Acrylamide with good hydrophility was copolymerized with A-PBLG through photo-induced polymerization and amphophilic PBLG-AM was successfully prepared. Results indicated that the hydrophility and biodegradation property of PBLG-AM was improved due to PAM introduced and crystalline degree of PBLG in PBLG-AM decreasing.A preliminary study of A-PBLG cross-linked with AM for drug delivery system was performed with model drugs of 5-fluorouracil(5-FU).It is found that the releasing rate and cumulative quantity of 5-FU increase with the increase of AM content,and decrease with the increase of cross-linker content at 37℃in PBS.
