Objective By carry out overall study of Dendrobium,To establish the inch decoctionpieces Dendrobium quality standards.Method TLC:Developing solvent Composed By Ethanol-butanone-acetyl acetone-water (15:15:5:85) Infrared detection wavelength with365nm. Microscopic identificationmethod: First to soften treatment, and then cut it to10~20μm, Put in the optical microscopeto make Microscopic Identification. Identification by Infrared Spectrometry:Take thepowder of Dendrobium candidum Wall.ExLindl,the water-soluble polysaccharide and mixedwith KBr respectively,tablet after grinding,test by infrared spectrometry.④Determination ofpolysaccharides:By use The Phenol-sulfuric acid method, use the D-anhydrous glucose asstandard,the Ultraviolet-visible spectrophotometry test and recovery test.⑤Determinationof the content mannose:uses the same method with Dendrobium medicinal,with anhydrousD-glucose, D-mannose as a reference,determined by HPLC. And repeatability test, standardprecision test, recovery test.⑥O ther Detection Methods:Refer to The first appendix Ⅸ ofpharmacopoeia of People’s Republic of China2010. Moisture: according to the first dryingH method. Total ash: According to the Determination of total ash K1method. Heavy metal:inspection method in accordance with Appendix E of the second method of heavy metal.Arsenic: in accordance with Appendix F, the first test for arsenic salt ancient Chua method.Extract: according to Appendix XA of Determination of alcohol soluble extract of hot-dipmethod.Results Results of microscopic identification are highly same with the standards ofDendrobium, TLC has a strong specificity; Infrared Spectroscopy was no significantdifference. Results of moisture, total ash, glucose, mannose and the peak area ratio ofheavy metals and arsenic salt Determination: Average water content8.28per cent; Averagetotal ash3.86per cent; mannose and glucose average area ratio4.38; heavy metals are notmore than20ppm; Arsenic content are not more than2ppm.Conclusions Dendrobium Medicinal herbs quality standards can be Applicable to the inchdecoction pieces Dendrobium,and make the limits for the different requirements.For heavymetals and arsenic salt, because levels is far lower than the minimum detection limit in the Pharmacopoeia(heavy metals:Not more than Twenty millionths, arsenic: not more than twomillionths).and then for this medicinal herbs is processed from cultivation, and the harmfulelements are easy to control in the cultivation process.so almost impossible toexcessive,Therefore, this limit is not included in the standard. In addition, InfraredSpectrometry Has the advantages of rapid, accurate,More and more commonly used intraditional Chinese medicine and herbs,Provides the reliable basis for the identification,soCan be added to the standard.
