Objectives:The objective of the experiment was to compare effects of treating cold syndrome of PD model rats by moxibustion SP6 with different intervention time. The experiment have two different intervention time,namely group A (moxibustion after model was established) and group B (moxibustion during model was being established). To compare effects of different intervention time of moxibustion on intensity of uterine contraction of cold stagnation syndrome dysmenorrhea model rats. Through molecular biology index of oxytocin receptor (oxytocin receptor,OTR), vasopressin receptors(vasopressin receptor,VPR),To investigate the part interference mechanism of different intervention time of moxibustion on relieving the pain of dysmenorrhea.Methods:64 dioestrum female rats were randomly divided into 4 groups, Respectively saline control group, model control group, group A, group B,with 16 rats in each group. In addition to the saline group, the rest of the experimental rats were given subcutaneous injections of estradiol benzoate injection for 10 days, the subcutaneous dose was 0.5 mg/each animal on the first day and the tenth day, and from the second to the ninth day,it was average 0.2 mg/each animal. After the last injection for 1 hour, each animal was given intraperitoneal injection of oxytocin for 2u, and placed into -25℃ freezer for 4 hours(with a 5 seconds interval between every 2 hours for ventilation) from 1-5 days after the injection. Animals in the saline group were injected with physiological saline of the same quantum. Animals in group A were treated by moxibustion on the 10th day after model was established while group B were treated by moxibustion on the 6th,8th,10th day. We use the BL-420-e biological function experiment system to detect intensity of uterine contraction in rats and rt-pcr method for detection of the mRNA expression of oxytocin receptor (OTR) and vasopressin receptors (VPR)in uterus tissue. Results:1. Effects of different intervention time of moxibustion on intensity of uterine contraction of cold stagnation syndrome dysmenorrhea model ratsCompared with the saline group, uterus contraction wave number, crest value and activity of model group rats were obviously increased or enhanced, the difference has statistically significance (P<0.01);Compared with model group, the number of uterine contraction wave and the activity of uterus of group A and group B both obviously reduced, the difference has statistically significance(P<0.01);There was no statistically difference in uterine contraction wave number, crest value and activity between the two groups (P>0.05).2. Effects of different intervention time of moxibustion on mRNA expression of OTR and VPR of uterus tissue of cold stagnation syndrome dysmenorrhea model rats(1) The mRNA expression of OTR:Compared with saline group,the mRNA expression of OTR of model group enhanced obviously,the difference has statistically significance (P<0.01);Compared with model group, the mRNA expression of OTR of group A and group B reduced obviously, the difference has statistically significance (P<0.01);and there was no statistical difference in the mRNA expression of OTR between the two groups (P>0.05).The mRNA expression of VPR:Compared with saline group, the mRNA expression of VPR of model group rised, but there was no statistically difference (P>0.05);Compared with model group, the mRNA expression of VPR of group A and group B reduced obviously, the difference has statistically difference (P<0.01);and there was no statistical difference in the mRNA expression of VPR between the two groups (P>0.05).Conclusions:1 Intensity of uterine contraction of cold stagnation syndrome dysmenorrhea model rats obviously enhanced, The cold stagnation syndrome dysmenorrhea model rats can be successfully prepared by the preparation methods of cold stagnation syndrome model combined with dysmenorrhea model.2 Early intervention of moxibustion SP6 and immediate intervention of moxibustion SP6 alone can both obviously reduce intensity of uterine contraction of the cold syndrome of primary dysmenorrheal rats, ease the pain state. The part analgesic mechanism of moxibustion is probably realized through inhibiting the abnormal mRNA expression of OTR, VPR in uterine tissue, thereby alleviating the state of abnormal and spastic contraction of uterine smooth muscle.3 There is no difference in moxibustion SP6 between early intervention and pure immediate intervention on cold stagnation syndrome dysmenorrhea model rats...
