| Purpose:Stroke is classically characterized as a neurological deficit attributed to an acute focal injury of the central nervous system by a vascular cause, including cerebral infarction, intracerebral hemorrhage, and subarachnoid hemorrhage, and is a major cause of disability and death worldwide. The burden of ischaemic and haemorrhagic stroke increased significantly between 1990 and 2010 in terms of the absolute number of people with incident ischaemic and haemorrhagic stroke(37% and 47% increase, respectively), number of deaths(21% and 20% increase), and Disability Adjusted of Life Years(DALYs)lost(18% and 14% increase). Stroke is a common problem in the process of aging population which cause social and medical problems in both developed countries and developing countries.In china, treatment of stroke is a hotspot of eye acupuncture therapy. Eye acupuncture therapy was developed by Doctor Jing-Shan Peng in the early 1970 s. His ideas came from classic Chinese medical sources and he divided the eye area into a total of four regions, eight areas, and thirteen pointsfor both the diagnosis and the treatment of disease. In recent years,eye acupuncture therapy is thought to be a kind of micro acupuncture because it is believed that the stimulations of the eye around the orbital margin can open the meridians, invigorate blood, stop pain, calm the Shen, and regulate the Zang Fu function. In China, the researchers are interested in the effect and the mechanism of the eye acupuncture and the relevance study was carried out by 973 program running by the Ministry of Science and Technology of the People’s Republic of China in 1999 the diagnosis of disease by observation of the eyes, treatment with eye acupuncture, the effect and its mechanism was carried out. Previous study collected clinical studies and trials to analysis the applications of eye acupuncture. Eye acupuncture therapy was used widely in clinical treatment including: cerebrovascular disease, pain, neurological disorders, mental disease.In this study we test the SOD MDA and GSH-PX Levels for cerebral ischemia reperfusion rats. Material and method:Inoxidizability processes may have a role in eye acupuncture. In this study we investigated erythrocyte superoxide dismutase(SOD) and,Glutathione peroxidase(GSH-Px);malondialdehyde(MDA) as a sign of Eye-acupuncture for Cerebral Ischemia Reperfusion Rats. 50 rats(SD,18 months,male) were used in this study and divided into 4 groups:control group, EA(eye-acupuncture) group,model group and pseudo group. We did normal breeding without any intervention in the control group;The string thrombus method was used to replicate the rat model of cerebral isehemia reperfusion injury in the EA group and model group then used eye-acupuncture therapy only in the EA group. The rats were sacrificed after 24 hours’ reperfusion with 10% chloral hydrate(3.5 ml/kg body weight) abdomen anesthesia, and The colorimetric method is adopted to detect MDA, SOD, gsh-px levels in serum. Results: 1. The mortality and neural function defect scaleAt the end of the therapy,the defect score were 0 points in both EA(eye-acupuncture) group and pseudo group. The score of MCAO model group was significant difference than the other three(p < 0.01). 2.The infarction volumeAs results of TTC stain, there were no infarcts occur in both control group and pseudo group. However, the rest of the two groups are infarcts. The infarct volume of MCAO model group was significantly higher than eye acupuncture group(p<0.01). 3.MDA, SOD and gsh-px levelsThe MDA content in serum of MCAO model group Groups of test results showed that of rats compared with blank control group and control group and eye acupuncture group were higher than control and pseudo model group significantly(p < 0.01);MCAO model control group and eye needle treatment group MDA content in serum of rats were significantly higher(p < 0.01), there is statistical significance; Compared with MCAO model control group, eye needle treatment group rats significantly reduce MDA content in serum(p < 0.01), with statistical significance. Compared with MCAO model control group, the leverof MDA was lower in the eye acupuncture group significantly(p < 0.01).Compared with blank control group and control group, the levels of SOD and gsh-px in serum was lower significantly(p < 0.01). Compared with MCAO model control group, the lever of SOD and gsh-px of eye acupunture group increased significantly(p < 0.01). Conclusion: 1 The eye acupuncture can increase the SOD level and reduce the MDA content in serum that might affect the cerebral ischemia reperfusion areas. 2 The eye acupuncture can improve behavior score and control infarcts effectively. 3The eye acupuncture probably achieve its antioxidant effect by increasing the content of SOD and gsh-px in serum and reduce MDA in serum. |
