Purpose:Based on NF-k B signaling pathway to explore "treating the AR acording to phlegm" of modern biology-based research by the effect of c AMP activator forskolin and PKA inhibitor H89, NF- kappa B hibitor PDTC and Xiaoqinglong Decoction and Yuping Feng powder on the expression of allergic rhinitis with AQP5,reveal rationality of the TCM "treating the AR acording to phlegm".Material and method:48 healthy male Wistar rats of clean grade according to the weight randomly divided into 6 groups, namely normal group, AR model group and PDTC intervention group(NF-k B inhibitor), H89(PKA inhibitor), Forskolin intervention group(c AMP activator) and Xiaoqinglong Decoction combined with Yuping Feng powder group. Each group was 8 rats. Except the normal control group, other groups were in ova and aluminum hydroxide as the sensitizing solution, allergic rhinitis rats by intraperitoneal injection of local attacking way. All groups were given corresponding drug intervention and after treatment, the rats were anesthetized rapidly after remove the nasal septum and the lateral wall of the nasal cavity respiratory mucosa tissue. Western blotting method was used to detect the expression of NF-k Bp65, p-CREB(Ser133) and expression of AQP5.Results:1 Rats in each group after treatment behavior comparison: forskolin and PDTC can improve the symptoms of allergic rhinitis. H89 can’t improve the symptoms of allergic rhinitis. Xiaoqinglong decoction combined with Yuping Feng Power group can alleviate the symptoms of AR rats and has the better treatment effect. 2 AQP5 Westem blotting results: AQP5 model group relative protein expression is significantly lower than the normal group. H89 treatment group AQP5 relative protein expression was significantly decreased compared with model group.Forskinlin treatment group, PDTC treatment group, Xiaoqinglong decoction combined with Yuping Feng Power treatment group AQP5 relative protein expression was significantly increased compared with the model group. 3 NF-kappa Bp65 Westem blotting results: In the model group, NF-kappa Bp65 relative protein expression levels were significantly higher than those of the normal group. H89 treatment group NF-kappa Bp65 relative protein expression was significantly increased compared with the model group. Forskinlin treatment group, PDTC treatment group,Xiaoqinglong decoction combined with Yuping Feng Power treatment group NF-kappa Bp65 relative protein expression was significantly decreased compared with model group. 4 p-CREB(Ser133) Westem blotting results: p-CREB model group(Ser133) relative protein expression is significantly lower than the normal group. H89 treatment group p-CREB(Ser133)relative protein expression was significantly decreased compared with model group. Forskinlin treatment group, PDTC treatment group, Xiaoqinglong decoction combined with Yuping Feng Power treatment group p-CREB(Ser133) relative protein expression was significantly increasedcompared with the model group.Conclusion: Xiaoqinglong Decoction and Yuping Feng powder can inhibit the activation of NF- kappa B signaling pathway, by combining the c AMP-PKA-CREB signaling pathway and CBP, incresed of AQP5 expression, thereby reducing the AR of rat nasal secretion. The expression of AQP5 is one of the TCM thought with Yang deficiency,the phlegm accumulation.
