Preliminary study on the nuclear import mechanisms of tumor metastasis suppressor gene 1Objective Tumor metastasis suppressor gene 1(TMSG-1/LASS2) is an important metastasis suppressor gene which is newly discovered. In normal and stimulated conditions, we detected the expression of TMSG-1 and apoptosis of Hela cells, and explored the nuclear import mechanism of TMSG-1 protein.Methods The expression of TMSG-1 was detected by immunofluorescence before and after H2O2 stimulation. After stimulation of different doses of H2O2, reverse transcription PCR and Western blot was used respectively to measure the expression of TMSG-1 m RNA and protein. Then we detected the different incidence of apoptosis by Flow Cytometry after H2O2 stimulation.Results Immunofluorescence results showed that the TMSG-1 expression in Hela cells could be detected both in natural state and stimulated state. The expression was much higher in stimulated state than in natural state. Reverse transcription PCR results showed that the TMSG-1 expression in Hela cells in stimulated state was much higher than that in natural state. With the increasing doses of H2O2, the TMSG-1 expression increased gradually. The obvious group was the H2O2 dose of 1.5m M. Western blot results also showed that the TMSG-1 expression in cells in stimulated state was significantly higher than that of natural state(P<0.05). The most obvious group was the H2O2 dose of 1.5m M.Flow cytometry results showed that there were different incidence of apoptosis before and after H2O2 stimulation(P<0.05), and the most obvious group was the H2O2 dose of1.5m M.Conclusion The TMSG-1 expression was significantly higher in cells of H2O2 stimulation than cells in natural state. After stimulation of quantitative doses of H2O2,TMSG-1 expression levels all increased, so as the cell number of apoptosis. Therefore,we conjectured that TMSG-1 nuclear import could be promoted by H2O2 stimulation,then TMSG-1 protein would promote cell apoptosis.To investigate the expression of tumor metastasis suppressor gene 1 in human prostate carcinomaObjective Tumor metastasis suppressor gene 1(TMSG-1/LASS2) is an important metastasis suppressor gene which is newly discovered. To investigate the expression of tumor metastasis suppressor gene 1(TMSG-1/LASS2)in human primary and metastatic prostate adenocarcinoma, and to explore its clinical significances, especially its role in tumor metastasis.Methods The expression of TMSG-1was detected in prostatic hyperplasia, primary and metastatic prostate adenocarcinoma immunohistochemically.Combining with tumor pathological behavior and clinical information,to explore the expression of TMSG-1 in human prostate cancer and clear its role in the process of tumor metastasis.Results TMSG-1 expression was observed in prostatic hyperplasia(20/22), primary prostate adenocarcinoma(34/38) and metastatic group(10/18). Difference was found between primary and metastatic group(P<0.05), and hyperplasia and metastatic group(P<0.05); but no difference between hyperplasia and primary group(P>0.05). TMSG-l expression was correlated with nerve invasion(P<0.05), while not correlated with Gleason grade, perirectal invasion and age(P>0.05, respectively) in primary prostate adenocarcinoma. Kaplan-Meier survival analysis showed that the survival time of patients was longer in TMSG-1 positive group than that in negative group(χ2=5.153,P=0.023).Conclusion TMSG-1 expressed higher in primary prostate adenocarcinoma than in metastatic one, indicating that the decreased TMSG-1 expression might promote cancer metastasis. Low expression of TMSG-1 can be used as a helpful risk factor for predicting metastasis of prostate adenocarcinoma. TMSG-1 may become a new target gene for clinical treatment and a reference index for prognostic evaluation of prostate adenocarcinoma.
