Objective: In order to illustrate the pathogenesis of aplastic anemia (AA) and investigate the therapeutic effects of cyclosporin (CsA) on AA. Methods: By using the methycellulose semisolid culture. We observed the changes of CFIJ-GM. BFU-E. CFU-E of bone marrow in 31 patients with AA. By using the APAAP method, We observed the changes of T-cell subgroup and HLA-DR antigen expression in the peripheral blood cells. CsA was administrated to 7 patients. Results: The colonies of CFU-GM. BFU-E. CFU-E wcrc strikingly decreased in AA patients. In T-cell subgroup, the CD44 ccli level dccrcascd CD8~ ccii level riscd, the CD4~/CD84 ratio decreased, their H LA-DR antigen expression rate was significantly higher than that of control. Four of seven patients were responsive. Conclusion: I.In the majority of AA patients, the hcmatopoictic stem/progenitor cells decreased and br defective, 2.the cell immune function was abnormal. 3. the CsA therapy was effective.
